摘 要:目的探讨雄性小鼠去势术后早期内环境变化特点,为合理有效建立前列腺良恶性动物模型在去势与非去势的选择方面提供参考。方法选用16只BALB/c雄性小鼠,随机平均分为去势组和对照组。去势组采用经阴囊途径双睾丸切除术,对照组则作同一切口假手术。第21天所有小鼠采血处死,分别检测血清睾酮(T)、雌二醇(E2)、血管内皮生长因子(VEGF)和SELDI蛋白分析。结果与对照组相比,去势组血清T接近成倍升高(730/400),E2反而成倍降低(112.53/256.45),T/E2较对照组升高约4倍(6.49/1.56);VEGF变化不明显(P>0.05);去势组在7个蛋白位点(3.4、5.3、7.1、11.6、11.8、15.2和15.9 KDa)有高表达,在2个位点(6.0和6.2KDa)为低表达。结论去势与否不仅对小鼠机体性激素、VEGF和蛋白表达等有影响,而且会导致在去势与否基础上所建立的前列腺良恶性模型特征不同及其研究结果差异。Objective To study early internal responsiveness after castration of male mice in order to provide some feasible consultations in model-building fields of prostatic benign and malignant diseases. Methods 16 male BALB/c mice were randomly distributed into two groups as castrated group and control group,8 mice in each group.Each mouse in castrated group was managed bilateral orchectomy through a scrotum incision,while those in control group were only accepted sham operations.All mice were sacrificed on the 21st day.Their prostates were studied by pathology while their serous testosterone,estradiol,VEGF and proteins were analyzed by ELISA and SELDI respectively. Results In castrated group,prostates became apparently atrophy and soft clothed with much more fat and fascia.Moreover,testosterone was nearly double increased(730/400) whereas estradiol was over double decreased(112.53/256.45),which correspondingly raised the ratio of T/E2 about 4 times(6.49/1.56).Nevertheless,VEGF had not distinct variance(P〉0.05).In addition,seven proteins(3.4,5.3,7.1,11.6,11.8,15.2 and 15.9 KDa) were in high expressions whereas two proteins(6.0 and 6.2 KDa) were in low expressions. Conclusions It is notable that not only inherent sexual hormones and proteins but also follow-up prostate benign or malignant models and the model-studying data would be probably influenced on the basis of pre-castration or not.
