用PCR方法定量检测IL-6受体基因在前列腺癌中的表达

摘　　要：利用逆转录聚合酶链反应（RT-PCR）定量检测了10例正常人前列腺、15例前列腺癌（PCa）及 PCa细胞系PC-3m中白细胞介素6受体(IL-6R）基因表达水平。结果发现,正常人前列腺组织中IL-6R基因呈低水平表达,PCa组织中IL-6R基因表达增强,明显高于正常组（P<0.01）,且73.3％（11/15）组织样本中IL-6R mRNA表达水平高于 PC-3m。而IL-6所介导的假单胞菌外毒素融合蛋白对该细胞系有强烈的杀伤作用。这表明IL-6及IL-6R与PCa的发生发展有关,并为PCa的生物学治疗的研究奠定了基础。By using RT-PCR tcchnque,IL-6R mRNA expression was quantitatively assayed in 10 normal prostate samples, 15 patients with prostate carcinomas (PCa) and a PCa cell line PC-3m. The results revealed that the expression level of IL-6R gene in PCa tissues was significantly higher than that in normal prostate,and 11 of 15(73. 3%)PCa samples expressed higher levels of an IL-6R mRNA than PC-3m which was sensitive to the cytotoxic action of an IL6-toxin fusion protein. Therefore,it is suggested that IL-6 may play a role in the growth of PCa,and IL-6R can be a target for the delivery of therapeutic agents on PCa.
