摘 要:目的研究不同浓度蜈蚣提取液(ECP)对肝癌(HCC)Bel-7404细胞株的敏感性。方法通过MTT实验、细胞药物浓度—时间生长曲线研究不同浓度ECP对Bel-7404细胞株的敏感性;利用光学与电子显微镜观察细胞形态及其超微结构变化。结果①MTT法揭示各浓度ECP对人HCC细胞Bel-7404生长均有抑制作用。②细胞生长曲线图显示Bel-7404活细胞数目与ECP浓度及药物作用时间相关。ECP在5 g/L时,Bel-7404活细胞数先增多,48 h后细胞数开始减少;浓度在10 g/L与20 g/L时,用药后24 h活细胞数就显著减少。各组间活细胞数比较有显著性差异(P<0.01)。③光镜下见ECP作用48 h后,浓度低于5 g/L对Bel-7404细胞无明显的抑制作用,浓度在10 g/L以上时,Bel-7404活细胞数减少,细胞形态改变,细胞坏死。电镜下见治疗组细胞密度增高,染色质凝集固缩,染色质边集,核固缩、核碎裂,及凋亡小体的形成。结论①ECP能抑制人HCC细胞Bel-7404的增殖,且抑制作用呈剂量和时间依赖效应;②ECP低浓度有诱导肿瘤细胞凋亡作用,中、高浓度有直接杀肿瘤细胞作用。Objective It is to study the sensibility of different drug concentration extract of centipede (ECP) on Bel- 7404 cell line of hepatocdlular carcinoma (HCC). Methods The sensibility of different drug concentration ECP on Bel- 7404 cell line was studied through MTT test and drug concentration-time growth curve. The cell morphons and the ultrastructural changes were observed through light microscope and electron microscope. Results ① MTT method revealed that different concentration ECP all had inhibitory action on the growth of HCC Bel - 7404 cell line. ② Cell growth curve chart showed that the viable counts of Bel - 7404 had relation with ECP concentration and drug action time. When the concentration of ECP was 5 g/L, the viable counts of Bel - 7404 increased firstly and then decreased gradually after 48 h. When the concentrations of ECP were 10 g/L and 20 g/L, the viable counts decreased significantly after medication for 24 h. There were significant dif- ferences on the viable counts among the groups (all P〈 0.01 ). ③ Observed through light microscope after medication for 48 h, ECP had no obvious inhibitory action on Bel - 7404 cell when the concentration was lower than 5 g/L. When the concentration was higher than 10 g/L, the viable counts of Bel- 7404 decreased, the cell morphous transformed and neorobiosis. Through electron microscope in treatment group, the cell density heightened, chromatic agglutination and pycnosis, chromatin margination, karyopyknosis, nuclear fragmentation and apoptotic body formation were observed. Conclusion ① ECP can inhibit the proliferation of HCC Bel - 7404. The inhibitory action is time-concentration dependent. ② Light concentration of ECP has the action of inducing tumor cell apoptosis. Middle and high concentrations of ECP has the action of direct killing tumor cell.
