摘  要:目的探讨DNA甲基转移酶抑制剂5-氮杂-2′脱氧胞苷(5-Aza-CdR)在骨肉瘤发生、发展中的作用及机制。方法采用不同浓度的5-Aza-CdR处理体外培养的骨肉瘤细胞Hs888T。MTT检测细胞增殖活性,流式细胞仪检测细胞周期及凋亡情况,Western blot法检测细胞Apaf-1蛋白表达水平。结果5-Aza-CdR处理后Hs888T细胞增殖明显受抑,细胞阻滞于G0/G1期,细胞凋亡增加,且与药物浓度及作用时间有关;Apaf-1蛋白表达水平显著增加。结论5-Aza-CdR在骨肉瘤发生、发展中起重要作用;可能机制为诱发Apaf-1在内的某些基因去甲基化。Objective To investigate the role and mechanism of DNA methylation transferase inhibitor 5-Aza-2'-deoxycytidine (5-Aza-CdR) in the occurrence and development of osteosarcoma. Methods Osteosarcoma ceils Hs888T cultured invitro were treated with different concentrations of 5-Aza-CdR. The proliferation activity of Hs888T cells were detected by MTr method, the cell cycle and apoptosis were observed by flow cytometry, the expression level of Apaf-1 protein was assayed by Western blot. Result After treated by the 5-Aza-CdR , the proliferation of Hs888T cells were significantly inhibited,the cell cycle were arrested in C0/C1-phase,the apoptosis increased in dose and time dependent manner,the expression level of Apaf-1 protein increased significantly. Conclusions 5-Aza-CdR play important role in the occurrence and development of osteosarcoma,which probably results from the demethylation of some genes as Apaf-1 .
