摘　　要：目的:研究Vcp mRNA表达与胃癌及淋巴结转移的关系,寻找术前预测淋巴结转移危险度及预后的标志基因。方法:采用RT-PCR、聚丙烯酰胺凝胶电泳EB染色法,检测42例胃原发癌、正常黏膜、淋巴结转移癌中Vcp mRNA的表达。结果:Vcp mRNA在上述组织细胞中均有表达,胃原发癌和淋巴结转移癌明显高于正常黏膜(P<0.01),且胃原发癌VcpmRNA表达与转移淋巴结数量具有相关性。结论:胃原发癌Vcp mRNA过度表达的患者淋巴结转移范围及程度相对较重,癌细胞侵袭力较强,宜行扩大根治术;术前RT-PCR检测胃原发癌Vcp mRNA表达有助于确定治疗方案及判断预后。Objective： To study the relationship between Vcp mRNA expression and gastric cancer and lymph node metastasis of gastric cancer, and to assess the significance of predicting the extent of lymph node metastasis and prognosis. Methods： Vcp mRNA expression was examined in matched primary lesions, normal gastric mucosa and lymph node metastatic lesions among 42 gastric cancer patients by reverse transcription polymerase chain reaction（RT- PCR）, polyacrylamide gel electxophoresis（PAGE）and EB stain. Results： The primary tumor samples showed a signifieantly higher level of Vcp mRNA expression compared with matched normal tissue（P 〈 0.01 ）. The Vcp mRNA expression showed a low level in benign lesions. The Vcp mRNA expression showed a higher level in patients with the number of lymph node metastasis. Conclusion： The degree of lymph node metastasis and invasiveness of carcinoma cells are more serious in cases with Vcp mRNA overexpression, and extensive lymph node dissection should be carried out for these patients. Examination of Vep mRNA expression by RT- PCR before surgery is indicated to estimate the prognosis.
