摘 要:[目的]观察硫酸镁预处理及硫酸镁治疗对兔全脑缺血后的神经保护作用。[方法]40只兔随机分为4组:对照组、缺血组、镁预处理组和镁治疗组,每组10只。阻断4血管(双侧颈总动脉及椎动脉),诱导全脑缺血,缺血时间为6min。测定缺血再灌注30min海马氨基酸含量。检测缺血再灌注3d海马CA1区神经元密度及凋亡神经元密度。[结果](1)镁预处理组及镁治疗组海马CA1区神经元密度显著高于缺血组(P﹤0.01),凋亡神经元密度显著低于缺血组(P﹤0.01)。(2)镁预处理组及镁治疗组海马天冬氨酸和甘氨酸含量显著低于缺血组(P﹤0.01);而γ-氨基丁酸含量显著高于缺血组(P﹤0.05)。(3)镁预处理组与镁治疗组比较各指标差异无统计学意义(P﹥0.05)。[结论]硫酸镁预处理及硫酸镁治疗对兔全脑缺血有显著的神经保护作用。[Objective] To observe the neuroprotection effect of magnesium sulfate pretreatment and magnesium sulfate therapy on global ischemia in rabbits. [Methods] Forty rabbits were divided randomly into three groups: control group (n = 10). ischemic group(n = 10). magnesium pretreatment group (n= 10) and magnesium therapy group (n = 10). The global ischemia was produced by block of four vessels (both carotid and vertebral arteries) for 6 min. The density of normal neurons and apoptotic neurons in the hippocampal CAI field were measured 3 days after reperfusion. The content of amino acid in hippocampus was measured 30 min after reperfusion. [Results] (1) The group and magnesium therapy group were significantly higher than that densities of normal of ischemic group neurons in magnesium pretreatment (P 〈 0.01), and the densities of apoptotic neurons in magnesium pretreatment group and magnesium therapy group were decreased significantly (P 〈 0.01 ). (2) The contents of asparfic acid and glycine in magnesium pretreatment group and magnesium therapy group were significantly lower than that of isehemic group (P 〈 0.01 ). The contents of GABA in magnesium pretearment group and magnesium therapy group were significantly higher than that of ischemic group (P 〈 0.05). (3) There was no significance between magnesium pretreatment group and magnesium therapy group (P 〉 0.05). [Conclusion] Magnesium sulfate pretreatment has significant neuroprotection effect on global ischemia in rabbits.
