摘  要:目的探讨趋化因子Mig、IP-10、I-TAC及其受体CXCR3的表达在大鼠肝移植手术前后的动态变化,分析其与肝移植急性排斥(AR)的关系。方法改良"二袖套"法建立大鼠原位肝移植模型和AR模型,分为4组:手术创伤组,肝移植无排斥组,肝移植急性排斥组,免疫抑制剂组。ELISA法检测血清Mig、IP-10、I-TAC表达。流式细胞仪检测外周血淋巴细胞CXCR3的表达,用Cellquest软件分析阳性细胞百分率。半定量RT-PCR检测各组第7天肝脏组织CXCR3-mRNA的表达。结果(1)各组大鼠手术后外周血趋化因子Mig,IP-10,I-TAC及其受体CXCR3表达出现明显升高。(2)AR组在术后第3~5天起Mig,IP-10,I-TAC及其受体CXCR3表达均高于其他各组(P<0.01)。(3)AR组大鼠在移植后第7天,外周血Mig、IP-10、I-TAC和CXCR3的表达随着AR组RAI积分的升高而逐步升高,相关系数分别为0.79,0.89,0.82,0.92(P<0.05)。结论血清中趋化因子Mig、IP-10、I-TAC及其受体CXCR3的高表达与AR密切相关,有望成为诊断AR较特异、敏感的指标。Objective To explore the dynamic expression of chemokines Mig,IP-10,I-TAC and their receptor CXCR3 around the time of acute rejection(AR) after liver transplantation in rats. Methods The models of orthotopic liver transplantation(OLT) in rats were established by two-cuff technique. Rats were randomed into 4 groups:Operation injury,OLT without AR,OLT with AR and OLT treated by FK506. The dynamic expression of chemokines Mig,IP-10,I-TAC and their receptor CXCR3 were measured 1 day prior to and 1,3,5,7 days after OLT. Results ( 1 ) On 3 - 5 days after OLT, the concentrations of chemokines Mig,IP-10,I-TAC in serum of rats in AR group were significantly higher than those of injury group, NAR group and FKS06 group (P 〈 0. 01 ). (2)The expression of CXCR3 in lymphocytes in the AR group was significantly higher than that of injury group and FK506 group 3 days after OLT and was significantly higher than that of NAR group 7 days after OLT ( P 〈 0.01 ). ( 3 ) The Mig,IP-lO ,I-TAC and CXCR3 in peripheral blood was significantly correlated with RAI on the 7th days after OLT,the correlation coefficients were 0. 79,0. 89,0. 82 and 0.92 ,respectively( P 〈 0. 05 ). Conclusion The dynamic expression of chemokines Mig, IP-10, I-TAC in serum and their receptor CXCR3 in the peripheral blood is closely related with acute rejection and could be used as a diagnosis indicator of acute rejection.
