摘  要:目的评价左旋聚乳酸(PLLA)和卵磷脂共混的新型复合材料的亲水性和细胞相容性,为血管组织工程(VTE)提供理想的支架材料。方法采用溶液共混法制备了PLLA/0—15%卵磷脂(质量百分比例)的共混物膜,通过测定静态水接触角评价不同材料的亲水性;将大鼠骨髓间充质干细胞(MSCs)接种在不同材料上以研究其细胞相容性,MSCs的生长增殖能力通过噻唑蓝(MTT)及乳酸脱氢酶实验检测,细胞形态通过碘化丙锭(PI)染色和扫描电镜观察。结果随着共混物膜中卯磷脂含量的提高,材料的亲水性能明显增加,MSCs在PLLA/5%卵磷脂共混物膜上增殖能力最佳且该材料没有明显的细胞毒性。结论PLIA/卵磷脂复合支架材料具有良好的亲水性和细胞相容性,可作为骨髓MSCs的理想的载体应用于血管组织工程。Objective To evaluate the hydrophilicity and cellular compatibility of novel PLLA/lecithin composite materials in order to provide optimal scaffold materials for vascular tissue engineering (VTE). Methods The blend films of poly ( L-lactic acid) (PLLA) containing 0-15% lecithin ( weight percent) were prepared by the solvent-casting technique. The hydrophilicity of different blend films was determined by measuring the static water contact angle. The cellular compatibility was studied by seeding rat bone marrow-derived mesenchymal stem cells (MSCs) on the films of PLLA containing 0-15% lecithin. Cell viability and proliferation were assessed using MTT and lactate dehydrogenase. Cell morphology was studied by propidium iodide staining and scanning electron microscopy. Results The hydrophilicity of PLLA was significantly increased with an increase of lecithin content in the blend films. 5% lecithin-containing PLLA films could possess higher adhesion and proliferation of MSCs, and didn' t demonstrate any significant toxic effects to cells. Conclusion The novel PLLA/lecithin composite materials have good hydrophilicity and cellular compatibility,which may be used as an optimal carrier matrix for bone marrow-derived MSCs in VTE.
