摘  要:目的:探讨腹腔镜结直肠癌手术人工气腹的安全性与可行性。方法:(1)实验分组为对照组,对照+用药组,CO2组,CO2+用药组,He组,He+用药组(PD98059);(2)用MTT法测定细胞增殖数;(3)用丫啶橙/溴乙啶(AO/EB)双荧光染色检测细胞凋亡率;(4)软琼脂克隆形成试验测定单个细胞增殖能力;(5)用免疫组化法测定结肠癌细胞中HIF-1α和VEGF的表达。结果:CO2气腹作用后人结肠癌细胞LS-174T增殖能力高于对照组、He组。He组细胞凋亡率高于CO2组和对照组,CO2、He人工气腹作用后结肠癌细胞中HIF-1α和VEGF阳性表达率高于对照组。PD98059药物组细胞生长速度、增殖能力及HIF-1α和VEGF的阳性表达率均低于相应处理组。结论:CO2气腹可促进结肠癌细胞LS-174T生长,He气腹可相对抑制其生长,PD98059可抑制结肠癌细胞的生长。Objective:To discuss the safety and feasibility of artificial pneumoperitoneum in the colon carcinoma operation and observe the influence of different pneumoperitoneal medium and PD98059 in the growth condition of colon carcinoma LS-174T cell in vitro. Methods : ( 1 ) experimentation grouping : control group, drug control group, CO2 group, drug CO2 group, He group, drug He group (uhimate PD98059 density in all drug group was 100umoL/L). (2)measurement of the cell proliferation condition by MTT. (3)detection of cell apoptosis ratio by AO/EB double fluorescein stain. (4) test of single cell proliferative capacity by soft agar clone formation experiment. (5)examination of the expression of HIF-1α and VEGF in colon carcinoma cell. Results: The growth speed and proliferating capacity of colon carcinoma cell LS-174T after CO2 pneumoperitoneum effect were higher than that of control group in vitro, while He group was significantly lower than the control group. Cell apoptosis ratio in He group was higher than that in CO2 group and control group. Positive expression of HIF-1α and VEGF in CO2 and He artificial pneumoperitoneum was higher than control group, and the expression in CO2 group was higher than He group, with both the significant difference. The cell growth speed, proliferation capacity, positive expression of HIF-1α and VEGF in PD drug group was lower than handled group with significant difference. Conclusions:CO2 pneumoperitoneum can promote the growth of colon carcinoma cell LS-174T,while He pneumoperitoneum and PD98059 can inhibit its growth relatively.
