摘  要:目的研究黄芪多糖对胃癌细胞的生长抑制及诱导凋亡作用。方法用RPMI1640培养液对低分化人胃腺癌细胞株MKN45、高分化人胃腺癌细胞株MKN28进行传代培养,取对数生长期细胞培养24h后分别予不同浓度的塞来昔布和黄芪多糖,空白阴性对照组只加培养液不加细胞。用噻唑蓝(MTT)法观察黄芪多糖及塞来昔布对胃癌细胞株MKN45和MKN28增殖的影响;用流式细胞术方法测药物作用后细胞凋亡和细胞周期变化。结果黄芪多糖和塞来昔布均可抑制胃癌细胞株MKN45和MKN28的增殖,效应呈浓度和时间依赖性,塞来昔布各浓度组加药24、48 h后,MKN45、MKN28均明显低于空白阴性对照组(P<0.05,P<0.01);而黄芪多糖5~20 mg/mL范围内加药24、48 h后MKN45、MKN28均明显低于空白阴性对照组(P<0.01),而黄芪多糖2~2.5 mg/mL浓度组加药48 h MKN28低于空白阴性对照组(P<0.05)。黄芪多糖可诱导MKN45细胞凋亡,阻滞MKN45细胞周期于G1期,但对MKN28细胞凋亡和细胞周期无明显影响;塞来昔布可诱导MKN45细胞凋亡,但对MKN28细胞凋亡及2种细胞的细胞周期均无明显影响。结论黄芪多糖能抑制胃癌细胞增殖,其机制之一是影响细胞周期使之阻滞于G1期,诱导细胞凋亡。Objective To observe the effect of inhibiting and promoting apoptosis of astragalus polysaccharides on gastric cancer cell. Method Subculture was carried on by poorly differentiated gastric adenocarcinoma cell line MKN45 and highly differentiated gastric adenocarcinoma cell line MKN28 in RPMII640 culture solution. After 24 hrs of culture logarithmic phase cell were treated with different concentration of astragalus polysaccharides and Celebrex. The cell cycle, apoptosis, and cell proliferation of both MKN45 and MKN28 were studied by flow cytometry ( FCM ) and MTY assay, respectively. Result Astragalus polysaecharides and Celebrex can inhibit cell proliferation of both MKN45 and MKN28. The inhibitory effect showed that a dose - , time - dependent manner. Proliferation of both MKN45 and MKN28 in different concentration Celebrex groups were lower than that in negative control group after 24hrs, 48hrs of adding drug ( P 〈 0.01 ). Proliferation of both MKN45 and MKN28 in 5 - 20 mg/mL astragalus polysaecharides groups were also lower than that in negative control group after 24hrs, 48hrs of adding drug ( P 〈 0.01 ). Proliferation of both MKN45 and MKN28 in 2 - 2.5 mg/mL astragalus polysaccharides groups were lower than that in negative control group after 24hrs, 48brs of adding drug ( P 〈 0.05 ). Astragalus polysaeeharides can induce apoptosis of MKN45, and block MKN45 cell cycle at G1 stage. There was no obvious effect on cell apoptosis and cycle of MKN28. Celebrex can induce apoptosis of MKN45. There was no obvious effect on cell apoptosis of MKN28 and cell cycle of both MKN45 and MKN28. Conclusion Astragalus polysaccharides can inhibit proliferation of gastric cancer cell. Influencing cell cycle and inducing apoptosis may be one of the mechanisms.
