散发性乳腺癌组织中BRCA1、BRCA2及P21^(waf)表达及临床意义

摘　　要：目的检测散发性乳腺癌组织中BRCA1、BRCA2及P21waf的表达,探讨BRCA1、BRCA2及P21waf在乳腺癌发生、发展中的作用。方法采用免疫组化Envision法检测90例散发性乳腺癌组织及35例乳腺腺病组织中BRCA1、BRCA2及P21waf蛋白的表达。结果BRCA1在乳腺腺病及散发性乳腺癌组织中的表达率分别为97.14%(34/35)、71.11%(64/90)。BRCA1与组织学分级、肿瘤大小、淋巴结转移情况及患者年龄无相关性。BRCA2在二者中的表达率为91.43%(32/35)、73.33%(66/90)。BRCA2与患者年龄有相关性,与组织学分级、肿瘤大小和淋巴结转移情况无相关性。乳腺腺病组织中不表达P21waf,在散发性乳腺癌组织中P21waf表达率为57.78%(52/90)。BRCA1与P21waf蛋白的表达无相关性(P>0.05)。结论BRCA1、BRCA2及P21waf蛋白异常表达可能在散发性乳腺癌的发生发展过程中起作用。Objective To investigate the roles of BRCA1, BRCA2 and P21^waf in the occurrence and development of sporadic breast carcinoma. Methods The immunohistochemistry method named Envision was used to detect the expression of BRCA1, BRCA2 and P21^waf in 90 cases of sporadic breast carcinomas and 35 cases of breast adenosis. Results BRCA1 protein expression rates were 97.14% and 71. 11 % in sporadic breast carcinoma and breast adenosis respectively. It had no significant correlation with histological grades, tumor size, lympho node status and patient age. The BRCA2 positive rates were 91.43% and 73. 33% respectively. The expression of BRCA2 was related to patient age. It＇s expression was not related to histological, tumor size and lympho node status. P21^waf was not found in breast adenosis. The positive expression rate of P21^waf was 57.78% in sporadic breast cancer. There was no significant relationship between the expression of BRCA1 and P21^waf （P〉0. 05）. Conclusions Aberrant expression of BRCA1, BRCA2 and P21^waf may play biological role in sporadic breast carcinoma developmerit.
