摘　　要：目的:探讨核因子-κB(NF-κB)在多发性骨髓瘤发病机制中的作用及其临床意义。方法:对2006年1月～2007年9月来本院就诊的26例初诊MM患者骨髓活检标本利用免疫组化方法检测NF-κB蛋白的表达,抽取2006年10月～2008年1月来我院就诊的17例初诊MM患者骨髓液利用RT-PCR方法检测NF-κB mRNA的表达。结果:初诊多发性骨髓瘤患者的NF-κBP65蛋白表达水平和对照组相比增高明显,蛋白主要表达于浆细胞的细胞核内;NF-κBP65 mRNA在初诊骨髓瘤患者表达强度高于对照组。NF-κBP65表达阳性的MM肿瘤负荷高、与多项临床指标有关。结论:本试验NF-κBP65在多发性骨髓瘤中高表达,可能参与疾病的发生发展,可作为预后参考指标之一。Objective： To investigate the clinical significance of nuclear factor-κB （NF-κB） in the pathogenesis of multiple myeloma （MM）. Methods：The bone man＇ow biopsy specimens from 26 newly diagnosed MM cases between Jan. 2006 and Sep. 2007 were detected for the expression of NF-κB by immunohistochemistry, and the bone marrow was extracted in 17 newly confirmed MM cases during Oct. 2006 and Jan. 2008 and determined by RT-PCR for the expression of NF-κB mRNA. Results： The protein expression levels of NF-κB p65 in the newly diagnosed multiple myeloma patients increased significantly as compared with the control group and the express was prominent in the nucleus of plasma cells. The higher NF-κB p65 mRNA was also found in the newly confirmed cases in comparison with the controls. The increased MM tumor load of positively expressed NF-κB p65 seemed associated with a number of clinical indicators. Conclusion：The higher NF-κB p65 expression in MM suggests that NF-κB p65 may be involved in the development of this disease, which can serve as one of the indexes for evaluating the prognosis.
