端粒酶和erbB4基因在非小细胞肺癌中表达的研究

摘　　要：目的研究端粒酶和erbB4基因在非小细胞肺癌(NSCLC)组织中的表达及与肺癌临床、病理的关系。方法应用TRAP法测定56例肺癌组织标本中端粒酶的表达。同时应用免疫组化法检测56例肺癌石蜡组织中erbB4的表达。记录56例肺癌患者5年生存情况。结果NSCLC组织中端粒酶和erbB4阳性表达率分别为75%(42/56)及89.3%(50/56)。端粒酶、erbB4表达与不表达在临床分期组间的差异有统计学意义(P=0.004、0.005),端粒酶表达与不表达患者5年生存率差异有统计学意义(P=0.0005)。结论检测端粒酶及erbB4表达有助于判断NSCLC临床特点及预后。Objectives To test the expression of telomerase activity and HER4 in non-small cell lung cancer and elucidate the relationship between the expression and the clinical pathology. Methods Telomerase activity in fresh specimens of lung cancer after ration from 56 lung cancer patients was detected by telomeric repeat amplification protocol （TRAP） and 56 cases of paraffin-embedded sues from the same specimens of lung cancer patients were tested for the expression of HER4 by means of Immunohistochemical assay. Results The positive rate of telomerase activity in NSCLC was 75 %（42/56） and HER4 was positively expressed in NSCLC in 89.3%/56）. The positive expressions of both telomerase activity and HER4 only correlated with the TNM staging （P=0.004,0.005）, and the positive expression of telomerase activity also correlated with the 5 year survival rate （P=0.0005）. Conclusions Telomerase activity and erbB4 may be useful as a diagnostic marker to elucidate the clinical characteristics and prognosis of lung cancer patients.
