摘  要:目的:研究加温联合六亚甲基二乙酰胺(HMBA)对人粘液表皮样癌MEC-1细胞的作用。方法:采用MTT法及软琼脂克隆形成法观察加温联合HMBA对MEC-1细胞的作用;并用免疫组化法检测加温及联合HMBA对多药耐药(MDR)表达蛋白(P-gp)的影响。结果:加温联合HMBA对MEC-1细胞有协同抑制作用。免疫组化染色显示,对照组及单用药组细胞P-gp中度表达;加温组细胞P-gp不表达;联合组为低度表达。结论:加温联合HMBA对粘液表皮样癌的治疗可能有一定价值。Objective:To study the effects of hyperthermia combined with hexamethylene bisacetamide(HMBA) on mucoepidermoid carcinoma MEC-1 cells. Methods:MEC-1 cells were treated with HMBA at various concentrations ,or hyperthermia at 42..5℃ for 1h,or of the combination of both.The inhibitory effects were investigated by MTT assay and colonogenic assey respectively.Expression of MDR protin P-gp was measured by immuno-histochemical staining. Results:1)hyperthermia could intensify the effects of HMBA; 2)MEC-1 cells expressed P-gp protein at a medium level , HMBA did not show effects on the expression ,but hyperthermia alone or with HMBA suppressed P-gp expression. Conclusion:Hyperthermia and HMBA may be potential in treating mucoepidermoid carcinoma.
