Influence of 5-HT on Circadian Electroactivity of Rat Suprachiasmatic Nucleus Neurons in Hypothalamic Slice

摘  要:采用脑薄片方法,观察损毁中缝背核(DR)、不同光照条件、5-羟色胺(5-HT)、氯氮平和利坦色林对视交叉上核(SCN)神经元昼夜自发放电的影响。结果:①SCN有昼高夜低的自发放电的节律性;②5-HT对SCN神经元自发放电有抑制作用,SCN神经元对5-HT的敏感性呈昼低夜高的节律(P<0.01);③氯氮平和利坦色林均能阻断5-HT对SCN神经元自发放电的抑制作用;④损毁DR后,SCN神经元对5-HT的敏感性增高,结果提示5-HT7样受体可能参与DR的5-HT能纤维对SCN神经元的调节作用。The influence of serotonin(5 - HT)、 clozapine、ritanserin and dorsal raphe nucleus(DR) Iesion on spontaneous firing of SCN neurons under various illumination conditions was examined using a hypothalamic slice preparation. It was found that:①SCN neurons persist circadian rhythm in spontaneous discharge. The firing rate of SCN neurons is high during subjective day and low during subjective night.②5 - HT inhibits the spontaneous firing of SCN neurons and its sensitivity to 5 - HT in subjective day is lower than that in subjective night(p<0.01 ).③Both clozapine and ritanserin can block the effect of 5 - HT.④The sensitivity of SCN neurons to 5 - HT is higher after DR lesion. The results suggested that the inhibitary effect of 5 - HT proection form DR on SCN neuron discharge might be mediated by 5 - HT7, like receptor.
