摘  要:目的:建立测定卵磷脂中5种主成分磷脂酰胆碱(PC)、磷脂酰乙醇胺(PE)、神经鞘磷脂(SM)、溶血磷脂酰胆碱(LPC)、溶血磷脂酰乙醇胺(LPE)含量的高效液相色谱-蒸发光散射检测器(HPLC-ELSD)法。方法:色谱柱为YMCPVA-Sil,以氯仿为流动相A、甲醇-水(96:4)为流动相B进行梯度洗脱,流速为0.5mL.min-1,柱温为25℃,检测器为ELSD,参数为漂移管温度40℃、载气(氮气)流量1.0bar。结果:PC、PE、SM、LPC、LPE进样量线性范围分别为2~12、2~12、0.5~3、0.5~3、0.25~1.5μg(r>0.9975);平均回收率分别为99.93%、99.82%、100.02%、99.03%、98.79%(n=9),RSD分别为0.66%、0.42%、0.55%、0.74%、0.98%(n=6)。结论:本法简便、快捷,结果准确可靠,可用于卵磷脂的质量控制。OBJECTIVE: To develop a simple HPLC- ELSD method for the determination of 5 major constituents ineluding phospbatidylcholine(PC), phosphatidylethanotamine(PE), sphingomyelin(SM), lyso phosphatidylcholine(LPC) and lyso- phosphatidytethanolamine(LPE) in lecithin. METHODS: It was separated on YMC PVA- Sil by gradient elution with chloroform as mobile phase A and methanol - water (96 : 4) as mobile phase B at a flow rate of 0.5 mL ·min ^-1; the column temperature was kept at 25 12 for separation. The detector was ELSD, of which the drift tube temperature was maintained at 40℃ and the flow rate of carrier gas N2 was kept at 1.0 bar. RESULTS: The linear ranges of PC, PE, SM, LPC, and LPE were 2-12, 2-12, 0.5-3, 0.5-3, and 0.25- 1.5 μg, respectively(r〉 0.997 5); their average recovery rates were 99.93%, 99.82%, 100.02%, 99.03%, and 98.79% (n = 9), respectively, and their RSD were 0.66%, 0.42%, 0.55%, 0.74%, and 0.98% (n = 6), respectively. CONCLUSION: The established method is simple, rapid with accurate and reliable results thus applicable for the quality control of lecithin.
