摘  要:目的探讨PTEN mRNA和蛋白在脑胶质瘤组织中的表达及临床意义,并分析其相关性。方法分别采用反转录PCR(RT-PCR)和免疫组化(I HC)方法检测PTEN在正常脑组织及不同组织学分级的人脑胶质瘤组织中的表达情况。结果Kruskal-wallis H检验显示,PTEN在脑胶质瘤组织中的表达与正常脑组织相比差异有统计学意义(P<0.01),且随着胶质瘤恶性程度的增高,PTEN表达强度逐渐下降。结论PTEN表达的缺失可能是脑胶质瘤发生、发展过程中极其重要的环节,PTEN可能是脑胶质瘤发生以及其预后判断的重要生物学指标。Objective To explore the clinical significance of the differential expression of PTEN in glioma tissue. Methods The mRNA and protein expressions of PTEN were assayed by reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry in 75 human brain glioma cases. Results The positive expression rate mRNA of PTEN differed significantly between brain glioma tissue and normal brain tissue ( Z2 = 22.66, P 〈0.01); the expression of PTEN was significantly higher in well-differentiated glioma than in poorly differentiated glioma. Along with the increase of glioma malignancy level, the expression intensity of PTEN decreased. Conclusion The decrease of the expression of PTEN mRNA and protein may have a very important effect on development and growth of glioma. It could be helpful in predicting the biological behavior and prognosis of glioma.
