摘 要:背景:幽门螺杆菌(H.pylori)为人类胃癌的Ⅰ类致癌原,但其在胃癌演进中的作用仍未完全阐明。目的:体外观察H.pylori对人胃癌细胞株AGS侵袭力的影响。方法:将AGS细胞与H.pylori标准菌株NCTC11637共培养72 h,光学显微镜观察12~72 h时细胞形态变化,扫描电镜观察24 h时细胞形态变化。侵袭小室实验检测共培养24 h后AGS细胞的侵袭力。结果:AGS细胞与H.pylori共培养12 h后,细胞渐出现变形,伪足增多、伸长,24 h和48 h时细胞间相互离散,伸出细长伪足,呈"蜂鸟"表型,60 h和72 h时细胞出现空泡样变,死亡逐渐增多;扫描电镜下可见H.pylori黏附于AGS细胞表面,细胞变形明显,伸出细长伪足。侵袭小室实验显示与H.pylori共培养24 h的AGS细胞,每200倍视野下穿膜细胞数显著高于PBS对照组(130.4±11.5对87.1±9.3,P<0.01)。结论:H.pylori感染可影响人胃癌细胞株AGS的细胞形态,增加细胞侵袭力。Background: Helicobacter pylori (H. pylori) is designated as class I carcinogen in human gastric cancer, however, its role in the progression of gastric cancer is not yet clearly known. Aims: To elucidate the effect of H. pylori on invasiveness of human gastric cancer cell line AGS in vitro. Methods: AGS ceils were co-cultured with standard H. pylori strain NCTC11637 for 72 hours. Morphological changes of AGS cells were examined with light microscope and scanning electron microscope, and the invasiveness of AGS cells was analyzed by transwell cell culture chambers. Results: Significant morphological changes and increased pseudopodium were observed in AGS cells after 12 hours coculturing with H. pylori. Hummingbird phenotype was observed at 24 hours and 48 hours, while increased vacuole formation and cell death were observed at 60 hours and 72 hours. Elongated pseudopodium of H. pylori attached AGS cells was also observed with scanning electron microscope at 24 hours. H. pylori infection rendered the AGS cells more invasiveness at 24 hours when compared with AGS cells incubated in PBS (cell crossing the membrane of transwell chamber, 130.4± 11.5 vs. 87. 1±9.3 per 200-fold field, P〈0.01). Conclusions: H. pylori infection induces morphological changes and enhances the invasiveness of human gastric cancer cell line AGS.
