摘 要:目的:研究牙龈卟啉单胞菌(Porphyrom onas gingivalis,Pg)膜表面蛋白HagA进行研究,探讨该蛋白在细菌黏附和入侵宿主细胞的过程中发挥的作用。方法:构建HagA全长基因到pYA292质粒上,并克隆到无致病性的沙门杆菌S.typhimuriumx4072菌中,构建hagA基因重复片段于pET19b质粒上,转入无致病性的大肠杆菌E.coliBL21中,检测hagA各段基因所表达的蛋白的功能。结果:S.tx4072 Hag A表达株对牙龈上皮细胞的黏附和入侵能力均明显提高,与空白对照组相比有显著性差异(P<0.05);E.coliBL21 HArep菌株仅提高了黏附力,对细胞的入侵能力与空白对照组相比无显著性差异(P>0.05)。结论:HagA全蛋白参与黏附和入侵牙龈上皮细胞的过程,HagA重复片段所编码的蛋白参与了细菌黏附细胞的过程。AIM:To determine the virulence of different parts of hemagglutinin A of Pg 381 in adhesion and invasion to human host cells. METHODS: The whole length of hagA gene of Pg 381 was cloned into the vector pYA292 in S. typhimurium x4072. Hemagglutinin repeat gene was cloned in vector pET19b and transferred into E. coli BI21. The function of different parts of the protein was tested for their ability of adhesion to and invasion of HGEC (human gingival epithelial cells, HGEC ) with a standard antibiotic protection assay. RESULTS: The experionental results showed that S. t x4072HagA increased the adhesion and invasion to HGEC in comparison to the control group with statistically significant difference (P 〈 0.05 ). The E. coil BL21Hrep improved the adhesion to the cells only. The invasion ability showed no statistically significant difference ( P 〉 0.05 ) in comparison with the controls. CONCLU- SION : These results suggest that HagA whole gene not only participates in adhesion but also take part in invasion of Pg to HGEC. The repeat unit of HagA might just participate in adhesion.
