肾性高血压大鼠脑缺血再灌注后脑组织NF-κB、ICAM-1、MMP-2和MMP-9表达的意义

摘　　要：目的观察肾性高血压大鼠脑缺血再灌注后脑组织中核转录因子(NF-κB)、细胞间黏附分子(ICAM-1)、基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶-9(MMP-9)表达的变化,探讨高血压加重缺血性脑损伤的机制。方法Wistar雄性大鼠20只,随机分为高血压组和正常血压组。前组采用狭窄肾动脉方法建立肾性高血压大鼠模型,正常血压组再分为假手术组(N)和缺血再灌注组(IR),分别行假手术和缺血再灌注处理。采用大脑中动脉线栓法造成大脑缺血再灌注模型,缺血时间为2h,再灌注22h。采用免疫组织化学技术检测脑组织NF-κBp65、ICAM-1、MMP-2和MMP-9的表达,用图像分析仪测定灰度值。结果与N组比较,IR组NF-κBp65、ICAM-1、MMP-2和MMP-9灰度值明显增高(P<0.01);与IR组比较,HIR组NF-κBp65、ICAM-1、MMP-2和MMP-9灰度值增高(P<0.05)。结论高血压加重缺血再灌注后脑组织NF-κBp65、ICAM-1、MMP-2和MMP-9的表达。Objective To observe the Expression of NF-κB,ICAM-1 ,MMP-2 and MMP-9 in renal hypertension rats with focal cerebral ischemia-reperfusion injury, and to research the mechanism of hypertension increased ischemic brain injury. Methods 20 male Wistar rats were randomly divided into two groups with hypertension and normal blood pressure group. The former group was treated with renal artery stenosis methods to perform rat model of renal hypertension. Normal blood pressure group was further divided into sham-operation group （N）and ischemia-reperfusion group （IR）, received sham surgery and ischemia-reperfusion treatment, respectively： Using the intraluminal suture method of middle cerebral artery to induce cerebral ischemia-reperfusion model of ischemia time was 2h and reperfusion 22h. The expression of NF-κBp65 , ICAM-1, MMP-2 and MMP-9 in the brain were detected by immunohistochemistry, and the gray value was measured by image analysis system. Results In the N group, IR group of NF-κBp65, ICAM-1, MMP-2 and MMP-9 showed significantly higher gray value （ P 〈 0.01 ） ; In the IR group, HIR group of NF-κBp65 , ICAM-1, MMP-2 and MMP-9 gray value increased （ P 〈 0.05 ）. Conclusion Hypertension aggravates ischemia-reperfusion brain tissue NF-κBp65, ICAM-1, MMP-2 and MMP-9 expression.
