摘　　要：目的建立多重PCR方法扩增外周T细胞淋巴瘤TCRβ链全长编码序列。方法根据现有文献,设计TCRβ链特异扩增引物。将退火温度接近的引物,组合配对进行多重PCR扩增并构建重组质粒,然后酶切鉴定。结果扩增出外周T细胞淋巴瘤外周血中的TCRβ链全长序列并成功构建重组质粒。结论建立了TCRβ链全长编码序列的多重PCR扩增方法。Objective To establish a method of Multi-PCR to amplify the complete DNA sequence of TCR β chains of patients with peripheral T-cell lymphoma.Methods The specific primers were designed according to literatures.Then to amplify the CDS of TCR β chain,and then inserted into cloning vector.The recombinant constructs were identified by the endonucleases.Results The CDS of TCR β chain were amplified,and constructed the recombinant plasmid.Conclusion In this study,a method of Multi-PCR to amplify TCR β chain were established.
