摘  要:目的:应用PCR-焦磷酸测序技术对食物中毒标本中沙门菌进行快速检测。方法:疑似沙门菌食物中毒的相关标本9份。前增菌后,进行核酸提取及PCR-焦磷酸测序技术检测。同时按GB/T4789.4-2008国标法进行细菌分离培养,利用VITEK全自动微生物鉴定仪对可疑菌进行生化反应检测,血清学试验鉴定血清型。结果:PCR-焦磷酸测序结果显示9份标本中3份沙门菌阳性。用分离鉴定方法也仅在该3份标本中分离出沙门菌。结论:PCR-焦磷酸测序技术可用于沙门菌食物中毒的快速检测。Objective:To apply PCR-Pyrosequencing technology on samples of food poisoning for rapid detection of Salmonella.Methods:After pre-enrichment of 9 specimens,PCR-pyrosequencing assay was used to detect Salmonella.At the same time,isolation and culture of bacteria were performed based on GB/T4789.4-2008.The use of automated microbial identification VITEK instrument carried out the detection of biochemical reactions for suspicious bacteria.Results:3 of 9 samples showed Salmonella positive by PCR-Pyrosequencing.3 strains Salmonella were isolated from 9 samples by traditional methods.Conclusion:PCR-pyrosequencing technology can be used as a rapid method for food poisoning of Salmonella.
