摘  要:目的研究丹参注射液对糖尿病大鼠脊髓背角神经元凋亡及cyclinD1表达的影响及其机制。方法成年雄性Wistar大鼠30只,随机分为糖尿病模型组(A组)、丹参治疗组(B组)和正常对照组(C组)各10只。A组和B组腹腔注射链脲佐菌素(STZ)60mg·kg-1建立糖尿病模型,分别于注射STZ前及注射后1,2,3,4,5周测定大鼠右后爪缩足反应阈值。B组机械性痛阈<8g开始腹腔注射丹参注射液3mL·kg-1,qd,连续4周。于注射STZ后5周处死大鼠,取L4~6脊髓,采用尼氏染色法光镜下观察脊髓背角组织病理学变化,TUNEL原位末端标记法检测脊髓背角神经元凋亡,免疫组化法测定脊髓背角神经元cyclinD1表达。结果与C组比较,A组与B组机械性痛阈降低,脊髓背角组织病理学损伤明显,神经元凋亡指数及cyclinD1表达均升高(均P<0.05);与A组比较,B组经丹参注射液治疗4周后机械性痛阈升高,脊髓背角组织病理学损伤减轻,神经元凋亡指数及cyclinD1表达均低于A组(均P<0.05)。结论丹参注射液对糖尿病神经病变有一定保护作用,能够减轻神经病理性疼痛,其机制可能与下调脊髓背角神经元cyclinD1表达、抑制神经元凋亡有关。Objective To investigate the effect of Radix salviae miltiorrhizae on apoptosis and cyclinD1 expression of spinal dorsal horn neurons in diabetic rats. Methods Thirty male Wistar rats weighing 180 -220 g were randomly divided into 3 groups (n = 10 each) : group A (diabetes mellitus) , group B (diabetes mellitus + radix salviae mihiorrhizae) and group C (normal control) . Diabetes mellitus was induced by intraperitoneal injection with streptozotocin (STZ) 60 mg·kg^-1 in rats. 3 mL ·kg^-1 Radix salviae miltiorrhizae was given intraperitoneally 1 week after STZ injection, q.d. for 4 weeks in group B. Paw withdraw threshold (PWT) was calculated by von Frey filaments before and 1,2, 3, 4 and 5 w after STZ injection. All rats were killed 5 weeks after STZ injection and lumbar segment of spinal cord was removed for detection of apoptosis in spinal dorsal horn neurons (TUNEL) and determination of cyclinD1 expression by immunohistochemistry. Results Compared with group C, PWT was lower, the histopathological damage in spinal dorsal horn was visible, apoptotic index and cyclinD1 expression were increased significantly in group A and B ( P 〈 0.05 ). While compared with group A, PWT was increased, the histopathological damage was attenuated, apoptotic index and level of cyclinDl were decreased significantly in group B ( P 〈 0. 05 ). Conclusion Radix salviae miltiorrhizae can attenuate the spinal dorsal horn neuronal apoptosis through down-regulation of cyclinD1 expression in diabetic rats.
