摘 要:目的:研究来曲唑诱导的多囊卵巢综合征(PCOS)模型大鼠卵巢组织中CYP19基因的表达,为该模型的应用提供一定的理论依据。方法:SD大鼠随机分为模型组和对照组,模型组应用来曲唑诱导大鼠PCOS模型。放射免疫法测定大鼠血清性激素水平;H~E染色观察卵巢组织学变化,免疫组织化学、Realtime-PCR和免疫印迹法检测CYP19基因在卵巢组织中的表达。结果:模型组血清性激素水平睾酮、黄体生成素、卵泡刺激素浓度显著增高,雌二醇、孕酮浓度降低。模型组CYP19mRNA及蛋白表达均显著高于对照组。结论:来曲唑诱导的PCOS大鼠卵巢组织中CYP19基因表达水平增高。Objective: To investigate CYP19 mRNA and protein expression in ovary tissue of rat model of letrozole-induced polycystic ovary syndrome (PCOS), and to provide experimental data for the model application. Methods: Forty rats were divided into two groups randomly, PCOS group administered letrozole once daily during 21 and a control group. Gonadal hormone concentrations in serum were determined by radioimmunoassay, the histologie changes in ovaries were observed by H-E staining, and the expression of CYP19 gene in ovary tissue was detected by Real time-PCR, Western blot and immunohistochemistry. Results: Compared to the control group, estradiol and progesterone showed a considerable reduction (P〈 0. 05). Levels of serum testosterone, FSH and LH in the model group were markedly increased than that in the control group (P%0.05). The expressions of CYP19 mRNA and protein were significantly up-regulated in the model group than those in the control group. Conclusion: The expression of CYP19 gene in letrozole-induced polyeystic ovary is significantly higher than that in the control ovary.
