摘  要:目的:利用培养的C2C12作为细胞模型,研究骨骼肌在电刺激强度相同,时间不同的情况下产生的活性氧与肌源性IL-6含量及其基因表达之间的关系。方法:以小鼠骨骼肌肌母细胞(C2C12)为研究对象,分为电刺激组和对照组。测定不同刺激时间C2C12肌管内活性氧(ROS)、IL-6mRNA表达以及C2C12肌管上清液中IL-6的含量。结果:1)电刺激各组的ROS、IL-6含量均显著高于对照组(P<0.05),峰值分别出现在刺激75min和120min;2)IL-6mRNA表达仅在刺激75min和120min时明显增加(P<0.05);3)各指标随电刺激时间的增加都有先增加,然后下降,至刺激90min时达低谷的变化规律,且ROS、IL-6含量在刺激90min时虽明显高于对照组(P<0.05),却明显低于其他各刺激组(P<0.05)。结论:1)C2C12肌管在电刺激下产生的IL-6浓度变化及IL-6mRNA表达均呈"双峰曲线",说明电刺激引起肌管IL-6蛋白变化可能与其基因转录的变化有关;2)电刺激引起C2C12肌管产生的活性氧与肌管IL-6蛋白含量和基因表达的变化规律一致,说明电刺激骨骼肌细胞收缩引起的活性氧的产生可能是肌源性IL-6产生和释放的调节因素之一。Objective:In this study, Ca C12 was used as the cell model, the relationship among reactive oxygen species, the contents of myogenic IL-6, IL-6 mRNA expression were researched in the same state of electrical parameter and time different. Method.C2C12 that differentiated 5 clays later was stimulated to contract in vitro, C2C12 cells were divided into stimulation and control groups (ES and CON groups). Reactive oxygen species (ROS), IL-6mRNA expression in C2C12 myotubes and the content of IL-6 in C2C12 myotubes supernatant were determined in the different stimulating time. Result : 1 ) with time of electric stimulation on C2C12 myotubes differed, ROS and IL-6 concentration of all the ES groups were significantly higher than the CON group (P〈0. 05). The peak value appeared at 75 min (P〈0. 01) and 120min (P〈0. 0l). 2) The IL-6mRNA expression in C2C12 myotuhes increased significantly at 75 min and 120min ES groups (P〈0.05 ). 3) All indexes had the same regularity with prolonged electrical stimulation, they first increased, then dropped to the bottom up to 90min. The ROS, IL-6 content in 90min were significantly higher than CON (P〈 0.05 ), but significantly lower than other ES groups (P〈0.05). Conclusion: 1) The change of IL-6 concentration and IL-6 mRNA expres- sion of rnyotubes with electrical stimulation were "double-peak curve". It showed the changes of IL-6 protein that caused by electrical stimulation might be related with the changes of IL-6 transcription. 2) The change regularity of ROS generated in C2C12 myotubes that caused by electrical stimulation was positively correlated with the change regularity of the protein level and gene level of IL-6. It showed that in the state of electrical stimulation, the generation of ROS was related with the generation and release of myogenic IL-6.
