摘  要:目的:研究β分泌酶抑制剂对岗田酸(OA)诱发PC12细胞损伤的保护作用。方法:将培养的PC12细胞分为3组:正常对照组、OA损伤组和药物保护组。OA损伤组用OA处理细胞,药物保护组在用OA处理前30min加入β分泌酶抑制剂。采用MTT法、免疫组化法观察β分泌酶抑制剂的保护作用,Western blot检测全长APP和β-C末端片段(β-C-terminalfragment,βCTF)。结果:药物保护组与OA损伤组相比细胞数显著增多,受损变圆的细胞数较少;OD490值显著升高(P<0.01)。OA诱导PC12细胞内全长APP和βCTF含量增加,β分泌酶抑制剂进一步增加细胞内全长APP含量,并减少βCTF含量。结论:β分泌酶抑制剂对岗田酸(OA)诱发PC12细胞损伤有保护作用,其机制可能和β分泌酶抑制剂减少βCTF水平有关。Objective:To study the protective effects of β - secretase inhibitor on injury of PC12 ceils induced by okadaic acid (OA). Methods:Culturing cells in medium with OA. β -secretase inhibitor as added in the medium. Morphological observation, MTT assays were used to study the protective effects of β -secretase inhibitor. APP, βCTF( β -C -terminal fragment)were determined by Western blot method. Results: β - secretase inhibitor attenuated round shaped cells induced by OA, increased OD490, Owing to OA treatment, APP and βCTF level increased in PC12 cells. β - secretase inhibitor increased APP level furthermore but decreased βCTF. Conclusion:β - secretase inhibitor prevents PC12 cells from injury by OA and the mechanism may be related to the decrease of the level of βCTF.
