摘 要:目的研究不同牙科烤瓷合金材料诱导细胞DNA损伤与细胞凋亡的情况,评价各种烤瓷合金材料的生物相容性。方法选择镍铬合金、钛合金、钴铬合金和金合金四种牙科烤瓷合金材料,制备四种合金材料的浸提液,用浸提液培养L929细胞24h。分别采用单细胞凝胶电泳(彗星试验)和流式细胞技术检测细胞DNA损伤与细胞凋亡的情况。结果镍铬合金、钛合金、钴铬合金可诱导不同程度的细胞DNA损伤和细胞凋亡(P<0.05),而金合金与对照组间无统计学差异(P>0.05)。结论不同烤瓷合金材料诱导细胞DNA损伤和细胞凋亡的情况存在差异性。金合金是种生物相容性优越的牙科烤瓷合金材料。Objective To investigate the DNA damage and apoptosis of L929 cells induced by different kinds of dental ceramic alloys and to evaluate the biocompatibility of alloys. Methods Leaching liquors of four kinds of alloy ( Ni-Cr alloy,Ti alloy, Co-Cr alloy and Au alloy) were prepared and used to culture the L929 cells. DNA damage of L929 cells was detected by the method of single cell gel electrophoresis (SCGE) and apoptosis of L929 cells was measured by flow cytometry (FCM). Results The DNA damage and apoptosis of L929 cells induced by Ni-Cr alloy, Ti alloy and Co-Cr alloy were different ( P 〈 0.05 ). There wasn' t significant difference between Au alloy and control group in the DNA damage and apoptosis of L929 cells ( P 〉 0.05 ). Conclusions Different dental ceramic alloys showed significant difference in the DNA damage and apoptosis of induced L-929 cells. Au alloy was a typical dental ceramic alloy material with good biocompatibility.
