摘 要:目的:研究大鼠激素性坏死股骨头内Runx2、Osterix基因表达和蛋白合成动态变化,探讨与骨合成代谢的关系。方法:用臀肌注射糖皮质激素和强迫负荷运动的方法制作股骨头坏死大鼠模型,HE染色确定模型诱导成功,提取大鼠股骨头内总RNA及总蛋白,行实时荧光定量PCR及Western blot检测,研究激素性股骨头坏死时股骨头内Runx2、Osterix基因表达、蛋白合成的动态变化。结果:造模8、10、12周时,模型组大鼠股骨头内Runx2和Osterix基因表达、蛋白合成较正常组减少,两者mRNA的表达量分别为正常组的0.9621、0.3259、0.0512和0.9661、0.2026、0.0194倍,且随时间推移两者的基因表达、蛋白合成呈下降趋势。结论:激素性股骨头坏死早期,股骨头内Runx2、Osterix mRNA表达和蛋白合成降低,抑制骨形成。Objective To observe the dynamic changes of gene expression and protein synthesis of Runx2(runt-related transcription factor-2)、Osterix and the influence of bone anabolic inside the femoral head of steroid-induced osteonecrosis in rats. Methods Early rat model of femoral head necrosis was made with alternate injection of dexamethasone bilateral gluteus maximus and training on laboratory animal treadmill.Hematoxylin and eosin stain,extract total RNA and total protein from the femoral head for real-time quantitative PCR,and Western blot were performed to detect the dynamic changes of the Runx2、Osterix genes expressions and proteins synthesis inside the femoral head of steroid-induced osteonecrosis in rats. Results At the post-modeling 8th,10th and 12th week,the expression of the Runx2 and Osterix genes,proteins synthesis in model group than the control group to reduce obviously,and the volume of mRNA expression were respectively equivalent to 0.9621,0.3259,0.0512 and 0.9661,0.2026,0.0194 times of those in control group.Both expression had a downward trend with the longer time. Conclusion Early steroid-induced femoral head necrosis in rats,the gene and protein expression of the Runx2、Osterix are inhibited,which inhibit bone formation.
