摘  要:为探讨对-壬基酚(p-nonylphenol,p-NP)摄入导致小鼠生精细胞损伤的机制,将野生型雄性小鼠分别用10%(V/V)p-NP腹腔注射7天和14天,应用苏木精-伊红染色、DNA片段化和Western印迹等检测方法观察p-NP干预后小鼠生精细胞损伤和凋亡的状况及其对Bcl-2和Bax表达的影响。结果显示,p-NP干预7天后,小鼠睾丸大体形态和睾丸组织细胞学形态均基本正常,但DNA片段化检测证明小鼠生精细胞出现DNA片段化的细胞凋亡典型表现;p-NP干预7天和14天后,C57小鼠睾丸组织Bcl-2表达强度下降,Bax表达增加,Bcl-2/Bax比值下降。提示p-NP对小鼠睾丸的毒性损伤机制之一是诱导生精细胞凋亡,bcl-2和bax基因参与了其引起小鼠生精细胞凋亡的调控过程。To explore the mechanisms of damage of testicular germ cells induced by injection ofp-nonylphenol (p-NP), wild-type C57 mice were injected with p-NP (10%, V/V) for 7 or 14 days. The damage and apoptosis of testicular germ cells was evaluated using methods of hematoxylin eosin staining, DNA fragmentation, and Western blot. The results showed that there was no morphological change in the epithelial cells of testicular tubules after 7- day p-NP injection, but more apoptotic germ cells were exhibited in these p-NP-injected mice than that in the wildtype mice. Furthermore, the mice subjected to a 7-day or 14-day injection of p-NP displayed a down-regulation of Bcl-2 and an up-regulation of Bax in the testis. These evidences suggest that the apoptotic regulation may be involved in the damage of testicular germ cells induced by acute p-NP injection, and bcl-2foax could contribute to regulating the apoptosis of testicular germ cells.
