摘 要:目的:研究青蒿素(artemisinin)对人宫颈癌HeLa细胞放射敏感性增强的作用机制。方法:MTT法检测青蒿素对正常人皮肤成纤维细胞GM0639、宫颈鳞癌SiHa细胞和宫颈腺癌HeLa细胞的生长抑制作用。克隆形成实验检测青蒿素对GM0639、SiHa和HeLa细胞的放射增敏作用。Western印迹法检测CyclinB1和Wee1蛋白在HeLa细胞中表达的变化。结果:MTT法检测结果显示,不同浓度的青嵩素(27.67~442.75nmol/L)对GM0639、SiHa和HeLa细胞作用24和48h时均有细胞生长抑制作用;而青蒿素(110.69nmol/mL)分别作用GM0639、SiHa和HeLa细胞6h后,对3株细胞的生长抑制率分别为0.93±0.25、0.84±0.10和0.77±0.03,对照组(未加药组)3株细胞的生长抑制率分别为0.92±0.13、0.83±0.09和0.75±0.21,加药组和对照组之间差异无统计学意义(P>0.05)。青蒿素对GM0639和SiHa细胞无放射增敏作用,对HeLa细胞有放射增敏作用,SER=1.12;青嵩素和照射联合作用HeLa细胞后,细胞中CyclinB1蛋白表达比照射组增加,Wee1蛋白表达比照射组减少。结论:青蒿素能增加HeLa细胞的放射敏感性,其作用机制可能与上调CylinB1及下调Wee1的蛋白表达有关。Objective:To investigate the effect of artemisinin in enhancing the radiosensitivity of human cervical cancer HeLa cells and elucidate the underlying mechanisms.Methods:The fibroblasts GM0639 ,cercival squamous carcinoma SiHa cells and cervical adenocarcinoma HeLa cells were cultured in vitro with artemisinin. The proliferation of cells was measured by MTT assay. Microco-lony formation test was used to determine the effects of artemisinin in enhancing radiosensitivity of GM0639,SiHa,and HeLa cells. Influence of artemisinin on related protein CyclinB1 and Wee1's expression levels was investigated by Western blotting in HeLa cells.Results:The inhibition rate of GM0639,SiHa,and HeLa cells were 0.93±0.25,0.84±0.10,and 0.77±0.03,respectively,after they were treated with artemisinin 110.69 nmol/mL for 6 h. The difference was not significant compared with control group 0.92±0.13 for GM0639 cells,0.83±0.09 for SiHa cells,and 0.75±0.21 for HeLa cells,P〉0.05). Artemisinin did not enhance the radio-sensitivity of GM0639 and SiHa cells but enhanced the radiosensitivity of HeLa cells. The SER was 1.12. The CyclinB1 level was increased but the Wee1 expression was decreased in artemisinin-treated HeLa cells compared with control cells.Conclusion:Artemisinin can enhance the radiosensitivity of HeLa cells. The mechanisms may be related with upregulation of CyclinB1 expression and downregulation of Wee1 expression.
