摘 要:目的探讨当归水提物及其活性成分阿魏酸(FA)对Caco-2细胞上的ABCB1蛋白摄取功能的影响。方法采用MTT法确定药物实验剂量,利用流式细胞术检测Caco-2细胞内ABCB1蛋白底物罗丹-明123(Rh-123)的荧光强度。结果由MTT法得到当归水提物和FA的浓度<100 mg.L-1时,细胞的存活率>90%;流式仪检测到中、高浓度的当归水提物和FA(10、100 mg.L-1)与Caco-2细胞孵育60 min后,分别能够使细胞内Rh-123的荧光强度减少27%、10%。结论当归水提物对ABCB1蛋白介导的Rh-123的外排有一定的诱导作用,并在一定范围内具有浓度依赖性,药物浓度越高,诱导作用越强。其活性成分FA对ABCB1蛋白只有轻微地诱导作用。Objective To determine the effect of water extract from traditional Chinese medicine Angelica sinensis and ferulic acid(FA) on the function of ABCB1 protein in Caco-2 cell monolayers.Methods Cell viability of Caco-2 cells with different concentrations of drugs was measured by monotetrazolium colourmetric assay,and we chose the appropriate concentrations of drugs for next steps.The fluorescence of rhodamine123(Rh-123) in Caco-2 cells was studied by flow cytometry.Results The cell viability of Caco-2 cells was higher than 90% when the concerntrations of water extract from Angelica sinensis and FA were less than 100 mg·L-1 with MTT.The water extract from Angelica sinensis and FA could respectively decrease the RH-123 intensity by 27% and 10% incubated with Caco-2 cells for 60 min.Conclusion The water extract from Angelica sinensis can moderately induce the function of ABCB1 protein-mediated efflux transport of Rh-123 in Caco-2 cells.The induction is concentration-dependent.FA shows slight induction to ABCB1 protein in this study.
