PAAF对大鼠血清TNF-α、内毒素及胃黏膜损伤的影响

摘　　要：目的探讨胰腺炎相关性腹水(pancreatitis associated ascetic fluid,PAAF)诱导的大鼠胃损伤作用及其相关机制。方法以PAAF诱导的SD大鼠胃损伤模型为研究对象,将32只SD大鼠(雌雄不拘)随机分为2组,每组16只。空白对照组,腹腔内注射生理盐水8ml/只;PAAF组,腹腔内注射PAAF8ml/只。于6h、12h分批处死大鼠,每时间点8只。测定血清内毒素和血清肿瘤坏死因子-α(tumour necrosis factor-α,TNF-α)的浓度,测定胃黏膜组织ATP酶的活性和TNF-α的浓度,于光镜下观察胃黏膜损伤情况。结果同时间点比较:PAAF组血清内毒素(LPS)、TNF-α、胃黏膜损伤程度和胃黏膜组织TNF-α的浓度,均较A组(空白对照组)增高(P<0.05),ATP酶活性显著降低(P<0.05)。结论PAAF能够引起大鼠胃黏膜的损伤。Objective To investigate the harm of the gastric mucosa induced by pancreatitis associated ascetic fluid （PAAF） in rats. Methods Thirty-two Sprague-Dawley （SD） rats, male or female, were randomly divided into two groups： blank group, 8 ml normal saline （NS） was injected into the peritoneal cavity; group PAAF as the model group, 8 ml PAAF was injected. After injection the rats were sacrificed in two batches at 6 h and 12 h, eight rats in each batch. Levels of endotoxin, TNF-α in serum were measured. The activity of ATPase and concentration of TNF-α in the gastric mucosal was measured. The pathological changes of intestines were observed under microscope. Results The levels of endotoxin, TNF-α in gastric mucosa and the degree of harm of the gastric mucosal, were much markedly elevated （P〈0.05）. The activity of ATPase of the gastric mucosa tissues was decreased （P〈0.05） in group PAAF than those in blank group. Conclusion PAAF can obviously induce the harm of the gastric mucosa.
