摘　　要：目的探讨抗血管生成药苏拉明(Suramin)对体外培养肝星状细胞(HSC)的影响及其抗纤维化的作用。方法将大鼠HSC-T6体外培养,用MTT法初测细胞增殖趋势、流式细胞仪定性及定量检测HSC的细胞周期和增殖指数;免疫组织化学检测α-平滑肌肌动蛋白(α-SMA);ELISA法测定细胞上清液中Ⅲ型胶原含量。结果与对照组相比,不同浓度Suramin组的HSC增殖均受抑制,阻抑在细胞周期G1期,呈时间和剂量依赖性(P<0.05)。各浓度组α-SMA灰度值亦低于对照组(P<0.05)。不同浓度Suramin组的上清液中Ⅲ型胶原均低于对照组,TGF-β1+Suramin组的含量介于TGF-β1组和Suramin组之间。结论抗血管生成药Suramin抑制HSC增殖和活化,从而减轻Ⅲ型胶原沉积,起到抗肝纤维化作用,即Suramin在临床应用上有多靶点抗肝纤维化的优势,但其具体机制有待进一步研究。Objective To observe the effects of Suramin on the rat hepatic stellate cell（HSC）-T6 in vitro and the effect of anti-fibrosis.Methods HSC-T6 in vitro culture were divided into a no-treatment control group and three different dosage Suramin-treated groups.The proliferation index and cell cycle of HSC were detected by MTT method and flow cytometry.The expression of α-smooth muscle action protein（α-SMA） was assessed by immunohistochemistry.The level of collagen type Ⅲ in the culture supernatant was quantitatively measured by ELISA.Results MTT method and flow cytometry showed that proliferation rates of HSC in Suramin groups were lower than that in control group,and the percentage of HSC in G1 phase increased significantly and proliferation index of HSC decreased significantly（P〈0.05）,the prevention effect was time and dose-dependent（P〈0.05）.The expression of α-SMA and the levels of collagen type Ⅲ in the Suramin groups were lower than those in the control group（P〈0.05）.The level of collagen type Ⅲ in the TGF-β1（10 μg/L）＋Suramin（1.2 g/L） group was higher than that in Suramin（1.2 g/L） group,but it was lower than that in TGF-β1（10 μg/L） group.Conclusion Suramin reduced the proliferation and activation of HSC,so as to reduce the aggradation of collagen type Ⅲ,which would prevent the development of liver fibrosis.Therefore,the multitarget-effects of Suramin on the anti-proliferation of HSC suggest a possible strategy of clinical application.
