摘 要:目的:研究miR-21在胃癌中的表达情况及其对胃癌细胞生物学行为的影响。方法:通过实时荧光定量PCR检测30例胃癌手术切除组织和周围正常组织中以及10株胃癌细胞中miR-21的表达情况。通过CCK8法、Annexin-Ⅴ+PI、流式细胞仪检测、划痕实验,观察过表达miR-21对胃癌细胞增殖、凋亡、细胞周期、迁移等生物学行为的影响。结果:定量real-time PCR结果显示miR-21在胃癌组织中高表达(P<0.01),流式细胞仪检测发现,miR-21过表达对细胞凋亡和细胞周期影响不明显(P>0.05),miR-21过表达细胞株增殖能力增强,与阴性对照组和空白对照组对比差别具有统计学意义(P<0.01),miR-21转染细胞划痕愈合速度较对照组快。结论:miR-21在胃癌中呈高表达,过表达miR-21可以促进BGC-823胃癌细胞的增殖能力和迁移能力,但对凋亡和细胞周期影响不明显。Objective To investigate the expression of miR-21 in gastric cancer and its influence on biological behavior of gastric cancer cell. Methods The expression of rniR-21 in 30 resected gastric cancer and its surrounding normal tissue specimens and 10 gastric cancer cell lines was measured by real time fluorescence quantitative PCR. The influence of overexpression of miR-21 on cell proliferation, apoptosis, cell cycle and migration were evaluated by CCK-8 kit, Annexin-V +PI, flow cytometry (FCM), and scratch healing assay. Results Real-time PCR showed overexpression of miR-21 in gastric cancer tissues (P〈0.01). Over-expression of miR-21 had no obvious effect on apoptosis and cell cycle (P〉0.05). Proliferation was enhanced in miR-21 overexpressed cell lines, significantly higher than that of negative controls and blank controls (P〈0.01).Tbe healing rate of miR-21 transfected cells was faster then that of controls in scratch healing assay. Conclusions miR-21 is overexpressed in gastric cancer. Over-expression of miR-21 promotes cell proliferation and migration, but has no obvious effect on apoptosis and cell cycle.
