摘  要:目的:研究细胞色素表氧化酶对在体肿瘤生长的影响。方法:将转染rAAV-CYP2J2、rAAV-CYPF87V、rAAV-antiCYP2J2和rAAV-GFP的Tca-8113细胞,分别接种至裸鼠皮下,观察皮下移植瘤的生长情况,并用免疫组织化学染色分析移植瘤细胞PCNA和CD31分子的表达情况。结果:转染rAAV-CYP2J2、rAAV-CYPF87V明显促进裸鼠皮下移植瘤的生长,并且使移植瘤出现时间(分别为5.8天和6.0天)明显较对照组和转染rAAV-GFP(分别为8.4天和8.6天)短;而转染反义rAAV-CYP2J2组移植瘤生长缓慢,出现时间也明显延长(约9.75天)。免疫组化结果显示,rAAV-CYP2J2、、rAAV-CYPF87V明显促进移植瘤细胞PCNA和CD31的表达,移植瘤微血管密度明显增加。结论:CYP表氧化酶能明显促进在体肿瘤的生长,其机制可能与促进肿瘤血管生成密切相关。Objective:To study the effects of CYP epoxgenase on growth of primary tumors in nude mice. Methods: Tca - 8113 cells infected with rAAV - CYP2J2, rAAV - CYPF87V, rAAV - antiCYP2J2 and rAAV - GFP were inoculated into nude mice, respectively. The growth curve of epidermoid carcinoma xenograft was evaluated. The expression of PCNA and CD31 molecule were detected by using immunohistochemistry. Results:Xenograft in the rAAV - CYP2J2 and rAAV -CYPF87V infection groups (5.8 and 6 days after injection) displayed significantly earlier than that in the non - infection control group and rAAV - GFP infection groups ( 8.4 and 8.6 days after injection, respectively) and much earlier than that in the rAAV -antiCYP2J2 group (9.75 days after injection). Immunohisto- chemistry analysis showed that infection of rAAV - CYP2J2, rAAV - CYPF87V significantly upregnlated the expression of PCNA and CD31 molecule and increased micro -vessel density in tumors and adjacent tumor interstitial tissues. Conclusion:These results provide a potent support to the conclusion that CYP epoxygenase is able to promote malignant proliferation of human cancers in vivo by increasing angiogenesis in tumors.
