摘 要:目的利用微透析法联合高效液相色谱法连续、在线、活体检测万古霉素在兔眼玻璃体内的浓度,探讨微透析法在眼科活体药代动力学研究中的可行性。方法将微透析探针植入兔眼玻璃体内24 h后,玻璃体内注射10 g.L-1万古霉素0.1 mL,利用微透析法直接采样,高效液相色谱法(HPLC-UV)连续检测30 min、1 h、2 h、4 h、6 h、12 h、24 h、48h、72 h和84 h正常兔眼和眼内炎兔眼玻璃体内万古霉素的浓度,并用计算机拟合并计算药代动力学参数。结果正常兔眼玻璃体内万古霉素的峰值浓度为(694.3±19.6)mg.L-1,半衰期为51.66 h;眼内炎兔眼玻璃体内万古霉素的峰值浓度为(721.4±29.4)mg.L-1,半衰期为11.91 h。所有的动物注药后84 h,玻璃体内万古霉素的浓度均高于最小抑菌浓度。结论微透析法是活体检测玻璃体内药物浓度的有效、直接的方法,可以直接反映出药物浓度随时间的变化,具有较高的可行性。Objective To investigate the feasibility of microdialysis in ophthal-mological pharmacokinetic studies by determining the concentration of vancomycin in vitreous of rabbit eyes by microdialysis and high performance liquid chromatography(HPLC) continuously,on line and in vitro.Methods A microdialysis probe was implanted in vitreous of rabbit for 24 hours,and 10 g·L-1 vancomycin 0.1 mL was administered intravitreally;The samples were collected with microdialysis,and the concentrations of vancomycin in vitreous of normal rabbit eyes and eyes with endophthalmitis continuously detected at 30 minutes,1 hour,2 hours,4 hours,6 hours,12 hours,24 hours,48 hours,72 hours and 84 hours by HPLC.Pharmacokinetic parameters were calculated with computer.Results In vitreous of normal rabbit eyes,the half life were 51.66 hours and the concentration of vancomycin at peak was(694.3±19.6)mg·L-1.In vitreous of rabbit eyes with endophthalmitis,the half life was 11.91 hours,and the concentration of vancomycin at peak was(721.4±29.4)mg·L-1.After intravitreally injected for 84 hours,concentration of vancomycin in vitreous was higher than minimal inhibitory concentration in all rabbit eyes.Conclusions Microdialysis is an effective and direct way to detect intravitreous drug concentration,and can reflect the changes of drug concentration with time.It has high feasibility.
