摘  要:目的:比较使用不同材料的染色容器对蛋白质十二烷基磺酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)银染显色速度及效果的影响。方法:以小鼠脑蛋白的SDS-PAGE为材料,凝胶分别在玻璃平皿、医用搪瓷盘和密胺塑料盘中进行银染,参照常规银染方法;同时采用一种优化的银染方法作为对照,在医用搪瓷盘中进行。银染后蛋白条带用Quantity One凝胶定量软件进行灰度值分析。结果:常规方法银染的凝胶,在玻璃平皿中显色最快,蛋白条带灰度值最高;在医用搪瓷盘中显色速度及灰度值均次之;在密胺塑料盘中显色速度及灰度值最低。对照优化方法银染的凝胶,显色速度和蛋白条带灰度值远高于常规银染的凝胶。结论:对于小鼠脑蛋白的SDS-PAGE银染,常规方法中,用玻璃平皿效果最好,医用搪瓷盘次之,密胺塑料盘最差。优化方法可以大大提高用医用搪瓷盘银染的效果。Objective:To compare the influences of application of different containers on developing speed and effectiveness of silver nitrate staining of protein in SDS-polyacrylamide gel. Methods:The mouse brain protein in SDS-PAGE was taken for material. The stained silver gels were carried out in accordance with routine silver staining method, in glass culture dish, enamel dish and melamine dish separately. Meanwhile an optimized silver staining method was used as control that was carried out in the enamel dish. After silver staining gray values of protein bands were analyzed by Quantity One gel analysis software. Results:The speed of developing the gels stained with routine method was the fastest. The gray values of protein bands were the highest in glass culture dish, secondary in enamel dish and the lowest in melamine dish. The developing speed and gray values of protein bands were much higher in control method of silver staining than that of routine method. Conclusion:In routine methods, the effectiveness of silver nitrate staining of protein in SDS-polyacrylamide gel was the best in glass culture dish, secondary in enamel dish and the lowest in melamine dish. The optimized silver staining method could improve the effectiveness using enamel dish.
