摘  要:目的:探讨人乳腺癌组织中过氧化物酶体增殖因子激活受体(peroxisome proliferators-activated re-ceptor gmma,PPARγ)和第十染色体同源丢失性磷酸酶-张力蛋白基因(phosphatase and tensin homology dele-ted on chromosometen,PTEN)蛋白表达的相关性。方法:采用免疫组化S-P法,检测20例人乳腺癌组织、癌旁组织中PTEN与PPARγ的表达情况。结果:PPARγ和PTEN的表达与乳腺癌患者年龄、肿瘤直径大小、TNM分期、病理学类型、孕激素受体(PR)及Her-2表达情况无显著相关性(P(0.05);肿瘤组织高分化组患者的PPARγ蛋白表达阳性率显著高于低分化组(P<0.05);腋窝淋巴结转移阴性者PTEN的表达显著高于≥4个淋巴结转移者(P<0.0125);雌激素受体(ER)阳性者PTEN的表达显著高于ER阴性者(P<0.05);癌旁组织PPARγ和PTEN的表达显著高于癌肿组织(P<0.01);PPARγ与PTEN蛋白在乳腺癌的表达成显著正相关(P<0.01)。结论:人乳腺癌中PPARγ与PTEN蛋白表达相关联,并可能参与乳腺癌的发生、发展、浸润与转移。Objective: To investigate the relevance between the expression of peroxisome proliferators -activated receptor gmma(PPARγ) protein and phosphatase and tensin homology deleted on chromosome ten (PTEN) protein. Methods: Detecting the positive expression of PPAR~ and PTEN protein in human breast cancer tissues and paracan- cerous specimens with SP immunohistochemical staining methods. Results: In human breast carcinoma tissues:there was no association between the positive expression of PPARγ and PTEN protein and the age of patients, size, TNM stage, histopathological type and the status of progesterone receptors or the expression of human epidermal growth factor receptor - 2 ( Her - 2) ( P 〉 0.05) ; the positive expression rate of PPARγ in the well - differentiated ( Grade l ) group was significantly higher than that in the poorly - differentiated ones( Grade Ⅱ,Ⅲ) (p 〈 0 0s) ;the positive expression rate in the patients with no one axillary lymph node metastasis was significantly higher than that with ≥4 ones( P 〈 0.01 ), with the PTEN expression rate in the estrogen receptor(ER) -positive group being significantly higher than that in the ER- negative group (P 〈 0.05 ) ;the positive expression rate of PPARγand PFEN in the paracancerous specimens of breast were both significantly higher than that in the breast cancer tissues(P 〈0.01 ) ;the expression of the two proteins were significantly positively correlated(P 〈 0.01 ). Conclusion: There is correlation between the expression of PPAR3, and PTEN protein in human breast cancer, and both of them may play a important role in the tumorigeness, development,infiltration and metastasis of human breast cancer.
