摘 要:目的探讨乳腺癌和乳腺良性病变组织中KAI-1 mRNA,Kiss-1 mRNA和MTA1的表达水平及其临床病理意义。方法检测60例乳腺癌和30例乳腺良性组织标本KAI-1 mRNA和Kiss-1 mRNA的表达情况。结果乳腺癌KAI-1 mRNA和Kiss-1 mRNA表达阳性率及其评分明显低于乳腺良性病变(P<0.05或P<0.01);乳腺癌MTA1表达阳性率及其评分均明显高于乳腺良性病变(P<0.05或P<0.01);导管内癌、组织学分级Ⅰ,Ⅱ级、无淋巴结转移、ER阳性及CA15-3阴性患者Kal-1 mRNA及Kiss-1 mRNA表达阳性率及其评分明显高于浸润性导管癌、组织学分级Ⅲ级、淋巴结转移、ER阴性及CA15-3阳性者(P<0.05或P<0.01),而MTA1表达阳性率及其评分则相反(P<0.05或P<0.01)。Kal-1 mRNA与Kiss-1 mRNA表达呈正相关(r=0.85,P=0.000);MTA1与Kal-1 mRNA及Kiss-1 mRNA两者表达均呈负相关(r=-0.64,r=-0.72,P=0.000)。Kal-1 mRNA和/或Kiss-1 mRNA表达阳性患者术后5年生存率及中位生存期均明显高于阴性者(P=0.000),而MTA1则相反(P=0.000)。结论KAI-1 mRNA,Kiss-1 mRNA和MTA1表达可能是反映乳腺癌发生、进展、转移或侵袭潜力及评估预后的重要生物学标记物。Objective To study the expressive levels of KAI-1 mRNA, Kiss-1 mRNA and MTA1 in the benign and malignant lesions of breast, and their clinicopathological significance. Methods We detected the expression levels of KAI-1 mRNA, Kiss-1 mRNA and MTA1 in the sections from 60 cases of breast cancer and 30 cases of benign lesions of breast. Results The positive rates and scores of KAI-1 mRNA and Kiss-1 mRNA were significantly lower in breast cancer than those in benign lesions( P 〈 0.05 or P 〈 0.01 ) ; but MTA1 were the opposite( P 〈 0. 05 or P 〈0. 01 ) . The positive rates and scores of KAI-1 mRNA and Kiss-1 mRNA were significantly higher in those with intra-ductal breast cancer, histological tumor grade I or II, non-metastasis of lymph node, positive expression of ER, and negative expression of CA15-3 than in those with ductal invasive breast cancer, histological tumor grade III, metastasis of lymph node, negative expression of ER, and positive expression of CA 15 -3 ( P 〈 O. 05 or P 〈 0.01 ) ; but MTA 1 expression was the opposite(P 〈 0.05 or P 〈 0.01 ). Highly close correlations were found among the expressive levels of Kal-1 mRNA and Kiss- 1 mRNA, but negative correlation between expression of MTA 1 and expressions of Kal- 1 mRNA and kiss- 1 mRNA in breast cancer tissues ( P 〈 0. 01 ). The Kaplan- Meier survival analysis showed the five-year survival rates and median survival times were significantly higher in the positive cases of Kal-i mRNA or Kiss-1 mRNA expression than those in the negative ones of Kal-1 mRNA or Kiss-1 mRNA expression in the breast cancer ( P 〈 0. 01 ) ; but those of MTA 1 were the opposite ( P 〈 0.01 ). Conclusions The expressive levels of Kal-1 mRNA, Kiss- 1 mRNA and MTA 1 might be important biological markers on the carcinogenesis, progression, biologic behaviors and prognosis of breast cancer.
