摘 要:为研究老鹳草提取物对人结肠癌QBC939裸鼠移植瘤组织中pcatenin和c—myc表达的影响,探讨老鹳草提取物对肿瘤发生、发展的作用,本研究成功建立了人结肠癌QBC939细胞株裸鼠皮下移植瘤模型,并将60只裸鼠随机分成空白对照组、老鹳草提取物组和羟基喜树碱组,每组20只,观察各组移植瘤体生长情况,免疫荧光法检测移植瘤组织中β-catenin和c—myc的表达。结果显示,与空白对照组相比,老鹳草提取物组和羟基喜树碱组移植瘤的体积明显缩小(P〈0.01),移植瘤组织中β-catenin和c-myc表达亦明显降低(P〈0.01);老鹳草提取物组移植瘤体积缩小及移植瘤组织中β-catenin和c—myc表达降低更明显(P〈0.01)。结果表明,老鹳草提取物可降低移植瘤组织中β-catenin和c—myc表达,对人结肠癌裸鼠移植瘤细胞具有抑制作用,且效果优于羟基喜树碱。The objective of this study was to investigate the effect of Geranium extract on the expressions of β-catenin and c-myc,thus to probe into the correlation of β-catenin and c-myc with the genesis and progression of tumour. Authors successfully established the hypodermic transplanted tumor model in nude mice by human colon carcinoma QBC939 cells, divided random the 60 nude mice into 3 groups,blank controlled group, Geranium extract group and Hydroxycamptothecine group, and observed the growth of transplanted tumor and determined the expressions of β-catenin and c-myc in tumor tissues. As results, compared with controlled group the contents of β-catenin and c-myc were decreased and the growth of transplanted tumor was inhibited in both Geranium extract group and Hydroxycamptothecine group significantly( P 〈0.01);in Geranium extract group the contents decrease and the inhibition on tumor were more significant( P〈0.01). Above results show that Geranium extract can reduce the expression of β-catenin and c-myc in the transplanted tumor tissues thus to inhibit the growth of tumor,it has better efficacy than Hydroxycamptothecine.
