摘 要:利用5L的玻璃抽滤瓶作为光生物反应器对发状念珠藻细胞进行开放及封闭式培养,考察了光强随细胞密度和光路长度的衰减变化。研究了光照强度、光照周期、pH、通气量对发状念珠藻细胞生长和胞外多糖产量的影响。结果表明,发状念珠藻生物量和胞外多糖产量均随着光照强度和通气量增加而提高。此光生物反应器能用于对发状念珠藻开放和封闭式培养,但封闭式培养更利于发状念珠藻生长及胞外多糖的产生。在光照强度60μmol·m-2·s-1,pH9.0,通气量0.7VVM的24h全光照条件下进行封闭式培养,生物量和多糖产量达到最高,分别为1.7g/L和89.32mg/L。本研究为采用低成本、易维护的简易装置进行发状念珠藻大规模培养提供了参考依据。Simple airlift photobioreactors were made using 5 L filtration bottles in which open and sealed cultures of Nostocflagelliforme were conducted and the influences of illumination intensity, photoperiod, pH, and ventilation on cell density and extracellular polysaccharides (EPS) were studied. The result showed that 5 L simple air-lift photobioreactor can be used for the open and close-end culture and both the biomass and polysaccharide production were improved with the increase of light intensity and ventilation in the culture. For the sealed culture, the best conditions were as follows: 0.7 VVM of ventilation, 60μmol·m^2·s^-1 of light intensity and constant illumination, under which the biomass and the polysaccharide production reached 1.7 g/L and 89.32 mg/L, respectively. This research provided some basic data for the large scale cultivation of Nostocflagelliforme with low cost, simple and easy maintenance equipment.
