肺癌中DAL-1表达及其与上皮细胞间质转化关系

摘　　要：目的探讨DAL-1在肺癌组织中表达及其与上皮细胞间质转化的关系。方法采用免疫组化EnVision两步法检测190例肺癌组织及163例癌旁组织中DAL-1、E-cadherin、Snail和vimentin表达,分析DAL-1与肺癌临床特征之间以及四种蛋白之间的相关性。结果肺癌组织中DAL-1、E-cadherin表达明显低于癌旁组织,差异有统计学意义(P<0.01);Snail、vimentin表达明显高于癌旁组织,差异有统计学意义(P<0.01)。DAL-1和E-cadherin表达降低与肺癌的淋巴结转移、临床分期、组织分化程度有关(P<0.01、P<0.05);Snail、vimentin表达升高与肺癌的淋巴结转移、临床分期、肿瘤大小有关(P<0.01)。DAL-1与E-cadherin阳性表达呈正相关、与vimentin阳性表达呈负相关(P<0.01);E-cadherin与Snail阳性表达呈负相关(P<0.01)。结论DAL-1可能参与了上皮细胞间质转化的过程,在肺癌淋巴结转移中发挥重要作用。Purpose To investigate the expression of DAL-1 in lung cancer and to explore its role on the epithelial-mesenchymal transition.Methods The expression of DAL-1,E-cadherin,Snail and vimentin was detected by immunohistochemistry in 190 cases of lung cancer tissues and 163 adjacent tissues samples.Analysis of DAL-1 protein and clinical features of lung cancer,as well as correlation among these proteins were conducted.Results Compared with adjacent tissues,lung cancer tissues had lower expression of DAL-1 and E-cadherin（P〈0.01） and higher expression of Snail and vimentin（P〈0.01）.Decreased expression of DAL-1 and E-cadherin protein was related to the lymph node metastasis,clinical stage,and degree of differentiation of lung cancer（P〈0.01 and P〈0.05）.The increased expression of Snail and vimentin was also correlated to lymph node metastasis,clinical stage,tumor size of lung cancer（P〈0.01）.The expression of DAL-1 was positively correlated with that of E-cadherin and negatively correlated with that of vimentin（P〈0.01）.E-cadherin expression was negatively correlated with that of Snail（P〈0.01）.Conclusions DAL-1 may be involved in the transformation of epithelial cells into the mesenchymal cells,and it may plays an important role in the lymph node metastasis of lung cancer.
