摘 要:目的:探讨白血病耐药细胞的发生机制与细胞内钙离子浓度的关系。方法:分别用Fura2/Am方法测定细胞内游离钙浓度及dot杂交法测定细胞mdr1mRNA表达。结果:发现CAs(Ver或Dil)有降低肿瘤细胞内Ca2+浓度和降低mdr1mRNA表达的作用,CaMAs虽无降低Ca2+浓度的作用,但可使mdr1mRNA表达降低,从而降低了K562/VCR细胞的耐药性。结论:细胞内Ca2+浓度的变化可能与MDR产生有一定关系;CAs导致的Ca2+浓度的变化可能也是其逆转肿瘤细胞MDR的机理之一。Purpose: To study the mechanism of the Multi drug resistant cells and the relationship to the calcium cation level in cells. Methods: the effect of CAs and CaMAs on intracellular Ca2+ concentation was determined and mdrl mRNA expression level of tumor cells with spectrofluorometry and Dot hybridization technique was used. Results: the results showed the calcium antagonists (Ver. and Dil) have the potential to decrease the calcium cation level within the tumor cells and the Mdrl mRNA expression. Although the CaMAs hasn't the potential to decrease the calcium cation level within cells, still has the potential to decrease the Mdrl mRNA expression thus lower the K562/VCR cell line's multi drug resistance. Conclusions: The change of intracellular Ca2+ concentration could be related to the production of MDR, the effect of CAs on intracellular Ca2+ may be one of the mechanisms of MDR reversal in cancer cells; CAs and CaMAs could also affect the mdrl mRNA expression.
