摘 要:目的和方法:采用NMDA(N-甲基-D-门冬氨酸,N-methyl-D-aspartate)受体NR1、NR2A及NR2B三种亚单位特异的抗体,对不同胎龄(12~33周)的人胚及成人脑皮层组织的这三种亚单位蛋白作了免疫印迹分析。结果:发现NR1、NR2A及NR2B三种亚单位蛋白在成人脑皮层组织均有表达;12周龄的人胚脑皮层组织就有NR1和NR2B亚单位蛋白的表达,随着胎龄增大此二种蛋白表达量随之增高,至33周龄时二者的含量达成人水平的75%左右;然而在上述所有人胚脑皮层样本中未能检出NR2A亚单位蛋白。结论:人胚脑皮层组织具有NR1和NR2B亚单位蛋白的表达。Abstract AIM and METHOD:By using quantitative immunobloting with NMDA receptor NR1, NR2A and NR2B subunit-specific antibodies, the protein levels of the three subunits have been determined in cortex tissues of human embryos aged between 12w~33w, as well as adult human cortex.RESULTS:NR1 and NR2B subunit proteins express in cortex of human embryos at least as early as 12 weeks of pregnancy, and the expressions of both the subunits have a same pattern of increase as embryos grow up. On the other hand, NR2A subunit protein is undetectable in all of the human cortex samples from the embryos aged between 12 to 33 weeks. CONCLUSION: Cortex of human embryos expresses NR1 and NR2B subunit proteins of NMDA receptor with increasing pattern at the developmental stage. MeSH Human;Embryo; Cerebral cortex; Immunobloting
