摘  要:目的:探讨耐药直肠癌细胞的耐药机制及中药逆转耐药机制。方法:在建立直肠癌药耐药细胞株的基础上利用流式细胞仪和Western-Blotting研究P-170、MRP及LRP等耐药蛋白的表达情况,以阐明耐药直肠癌细胞的耐药机制,另外用中药丹皮酚进行多药耐药的逆转实验。结果:LOVO/氟尿嘧啶耐药细胞株耐药机制主要是通过上调P-170、MRP等耐药蛋白的表达实现,而中药丹皮酚则可以通过下调P-170、MRP等耐药蛋白的表达来逆转该细胞株的耐药性。结论:对直肠癌耐药及相关逆转机制的研究有助于开发新的直肠癌治疗药物。Objective: To investigate the resistance mechanism of drug-resistant rectal cancer cells and Chinese medicine to reverse drug resistance mechanisms. Methods: On the base of establishing rectal cancer drug resistance cell line, study P-170, MRP and LRP by using flow eytometry and Western-Blotting to elucidate the mechanism of drug resistance, and using traditional Chinese medicine paeonol reversal of muhidrug resistance experiments. Results: LOVO/fluorour-acil resistance in the mechanism of drug-resistant mainly by increasing P-170, MRP protein expression.Chinese medicine paeonol could be adopted to reduce P-170, MRP protein expression to reverse the resistance cell lines' drug resistance. Conclusion: Investigate mechanism of rectal cancer drug resistance and its retroconversion will help to develop new cancer drugs.
