摘 要:目的应用L5178Y细胞检测芥子气(Mustard gas,MG)诱发tk位点突变。方法采用96孔微孔板接种法检测平板接种效率和突变频率。结果芥子气诱发了L5178Y3.7.2-tk+/-细胞tk位点的突变,诱发突变是自发突变的2~15倍。在tk位点诱发了大克隆和小克隆两种不同表型的突变集落,但以小克隆为主。结论L5198Y细胞检测MG诱发非位点突变可应用于化学战剂诱变性的检测,为tk基因突变试验评价化学战剂诱变性提供了实验方法。Objective To apply the method of tk gene mutation assay in LS178Y3.7.2-tk^+/- mouse lymphoma cells for testing gene mutation induced by mustard gas. Method The plating efficiency and the mutant frequency were detected by the method of 96 micro wells. Result Mustard gas induced tk locus mutation with mutation frequency 2-15 times higher than that of spontaneous mutation frequency of LS178Y3.7.2-tk^+/- cells. There were two different phenotypes of mutation colonies: large and small colonies, but most of them were small colonies. Conclusion Mustard gas induced tk locus mutation in LS178Y3.7.2-tk^+/- cells. MLA (mouse lymphoma assay) can be used in mutagenesis test of chemical warfare agents.
