摘  要:目的观察凝血酶对人肝癌细胞株HepG2增殖的影响及其诱导凋亡的作用。方法体外培养人肝癌细胞HepG2,MTT法观测不同浓度凝血酶在不同时间点对肝癌细胞增殖的影响,TUENL法检测其诱导凋亡作用。结果高浓度凝血酶对肝癌细胞增殖有明显抑制作用(P<0.05),当浓度大于100U/ml作用时间在48h后,其抑制率最高可达到(39.08±2.32)%,且不随浓度的提高和时间的延长而增加;高浓度凝血酶能够诱导肝癌细胞凋亡,当浓度大于100U/ml且作用时间大于48h后,各组凋亡指数无显著性差异(P>0.05)。结论高浓度凝血酶能够明显抑制体外生长的人肝癌细胞株HepG2并诱导肝癌细胞凋亡,且存在一定时间(48h内)及浓度(﹤100U/ml)依赖性。Objective To investigate the effect of thrombin on the proliferation and apoptosis of human hepatocellular carcinoma cell line HepG2. Methods HepG2cells were treated with thrombin at the doses of 0, 25, 50, 100, 150, or 200 U/ml for 24, 48 and 72 h respectively. The proliferation of the cells in each groups was measured by MTT assay. Cell apoptosis was analyzed with TUNEL assay. Results Higher doses of thrombin had obvious inhibitory effect on the proliferation. When the dose was increased to 〉 100 U/ml and treated for 48 h, the inhibitory ratio of the HepG2 cells was raised to (39.08 ± 2.32) % , which were not further elevated with the increase of dose or elapse of treatment time. HepG2 cells apoptosis was induced by thrombin, in a dose- and time-dependent manner when the dose was between 25 to 100 U/ml. However, there was no significant difference in the apoptosis of different groups when the dose was higher than 100 U/ml and the time exceed 48 h. Conclusion Higher doses of thrombin can inhibit the proliferation of HepG2cells, and lead to cell apoptosis. The both effects are increased in a dose- and time-dependent manner when the dose is 〈 100 U/ml and treatment time is within 48 h.
