摘 要:为观察青春期大鼠接触雌激素如辛基酚(OP)或三羟异黄酮对乳腺癌中NF-κB的影响,通过免疫组织化学、RT-PCR、Western blotting和电泳迁移率改变实验(EMSA)等方法观察雌激素影响大鼠乳腺癌中NF-κB P65的改变.结果表明NF-κB P65在细胞胞浆和胞核中均有表达,在乳腺癌组织中以胞核表达多见.与模型组(mod)比较,给予40 mg/kg辛基酚(OP40)或500 mg/kg三羟异黄酮(GEN)处理的大鼠乳腺癌中NF-κB P65 mRNA表达明显降低,且细胞核中NF-κB P65蛋白表达也明显减少,NF-κB DNA结合活性明显降低.OP及GEN降低大鼠乳腺癌发病可能与减少NF-κB P65核转位、降低NF-κB的DNA结合活性有关.To observe the effects of NF-κB expression when prepubertal rats were treated with estrogens such as octylphenol (OP) and genistein (GEN). By using the methods of immunohistochemistry, RT-PCR, Western blotting and electrophoretic mobility shift assay (EMSA), we have measured the changes of NF-κB expression in prepubertal rats treated with estrogens. Results showed that NF-κB P65 expression existed in kytoplasm and nueelus and that there were more NF-κB P65 expression of nucelus of mammary cancer tissue. Compared with the model group,the NF-κB P65 mRNA expression decreased significently in mammary cancer tissues when rats were treated with 40 mg/kg OP (OP40) or 500 mg/kg GEN (GEN) ,and NF-κB P65 expression in nucelus also decreased obviously.NF-κB DNA binding activity decreased significently in OP40 or GEN group when compared with the model group. Our conclusion was that the reduction of mammary cancers developed in OP and GEN groups was possibly related to the lowering down of NF-κB P65 nuclear translocation and low level DNA binding activity.
