端粒酶逆转录酶的表达与子宫颈癌前病变进展的关系

摘　　要：目的通过检测宫颈癌前病变组织中端粒酶逆转录酶(human telomerase reverse transcriptase,hTERT)的表达,探讨其与宫颈癌前病变进展的关系。方法采用免疫组织化学法检测52例宫颈组织病理存档石蜡包埋组织hTERT的表达,阳性信号采用图像分析仪进行定量分析。结果慢性宫颈炎组及CINⅠ组、CINⅡ组、CINⅢ组中的hTERT平均光密度值(AOD)分别为:(0.105±0.024)、(0.128±0.018)、(0.137±0.021),(0.201±0.016)。hTERT在各组表达的平均光密度值依次为CINⅢ级>CINⅠ、Ⅱ级>慢性宫颈炎,差异有统计学意义(P<0.05)。结论hTERT的表达与宫颈癌前病变进展相关,采用免疫组织化学法检测hTERT可能成为宫颈癌标志物,追踪hTERT阳性的子宫颈癌前病变患者,可能有助于早期发现宫颈癌。[ Objective ] To investigate the expression and the significance of telomerase hTERT in cervical precancerous lesions. To analyse whether a relationship between expression of telomerase hTERT and progression of cervical precancerous lesions. [Method] hTERT expressions in 52 cases of CIN and chronic cervicitis were detected by immunohistochemistry method and automatic image analysis technique. [ Results ] The A values of hTERT in the chronic cervicitis group, CIN Ⅰ group, CIN Ⅱ group and CIN Ⅲ group are separately： 0.105±0.024,0.128±0.018,0.137±0.021,0.201±0.016. The expression of hTERT in CIN Ⅲ group is significantly higher than that in other groups （P 〈 0.05）.The expression of hTERT in CIN Ⅱ and CIN Ⅰ are significantly higher than that in the chronic cervicitis group （P 〈0.05） between which there are no differences （P 〉0.05）. With the development of cervical lesion,the expressions of hTERT was increasing. [Conclusions] The expression of hTERT is associated with the progression of cervical intraepithelial neoplasias. Increased hTERT expression might be a useful marker for diagnosis of cervical carcinoma and an effectively tool for predicting the malignant change potential of cervical precancerous lesions.
