摘 要:目的研究结肠癌的发生机制。方法以级联多聚酶链式反应及逆转录定量多聚酶链式反应对21例结肠癌和近切端结肠组织,以及可获得的15例相应癌旁组织进行细胞凋亡调控基因bcl-2t易位和C-myc、mRNA转录水平检测。结果 bcl-2基因易位率和C-myc基因mRNA转录水平在结肠癌与正常组织、癌旁组织与正常组织均有显著性差异,而结肠癌与癌旁组织间无显著性差异,二者与Dukes分期间无显著相关性。结论 bcl-2基因易位和C-myc基因mRNA转录水平增高均为结肠癌发生的早期分子事件。bcl-2基因和C-myc基因的变化协同作用,抑制细胞凋亡,在结肠癌发生过程中起到重要作用。Objective To study the pathogenic mechanism of colonic carcinoma. Methods Nested polymerase chain reaction (N-PC.R) and reverse transcription quantitative polymerase chain reation (RT-Q-PCR) techniques were used to detect the translocation of apoptosis regulating gene bcl-2 and mRNA transcrip-tion level of C-myc gene in 21 samples of tumor and proximal colonic tissue, and 15 samples of corre-sponding tissue surrounding the tumor collected from the patients with colonic carcinoma. The correla-tion between the data and the Dukes stage of colonic carcinoma was analyzed. Results The transloca-tion rate of bcl-2 gene and mRNA transcription level of C-myc gene showed significant difference be-tween colonic carcinoma and normal tissue, or tissue surrounding the tumor and normal tissue; In con-trast, no significant difference was found either between colonic carcinoma and the tissue surrounding the tumor or between the alteration of the genes and Dukes' stage. Conclusion The translocation of bcl-2 and the increase of mRNA transcription level of C-myc may be the early molecular events in occur-rence of colonic carcinoma. The synergistic effect of bcl-2 and C-myc genes may inhibit apoptosis and play an important role in the pathogenesis of colonic carcinoma.
